Patients diagnosed with GBM are treated in a standardized manner that includes surgical resection and radiochemotherapy, mostly with the aggressive alkylating agent temozolomide. However, these patients present a very poor clinical outcome, with a median overall survival of approximately 15 months. Moreover, the high heterogeneity displayed by GBMs contributes to the patients’ poor response to therapy, making them still an incurable disease. The identification of novel prognostic biomarkers may help to achieve a more accurate prognosis and to perform a better patient stratification that could potentially open the possibility for more personalized therapeutic strategies. Cadherins are calcium-dependent cell adhesion molecules whose implication in cancer has been widely studied. P(Placental)-cadherin encoded by the CDH3 gene is a classical cadherin reported both as a tumor promoting or suppressor molecule, depending on the tumor type and the molecular context. Considering that nothing is known about P-cadherin in gliomas, we aimed to understand the functional, mechanistic and prognostic implications of CDH3/P-cadherin in gliomas, and more specifically in GBM. Our data indicate that CDH3/P-Cadherin is overexpressed in a subset of high grade gliomas at RNA and protein levels, suggesting P-cadherin may have a role in the aggressiveness of gliomas. Upon evaluating the functional impact of CDH3/P-cadherin in GBM using silencing and overexpression in vitro models, we found that P-cadherin regulates various cancer hallmarks in GBM, being associated with increased cell viability and cancer stem cell properties. Additionally, P-cadherin also impacted the invasion capacity of GBM cells. Noteworthy, the oncogenic roles observed in vitro were validated in vivo using an orthotopic mouse model of GBM, as animals injected with GBM cells overexpressing CDH3 displayed a significantly shorter overall survival. Mechanistic analyses showed that CDH3 positively correlates with genes involved in cancer-related signaling pathways. Importantly, the prognostic value of CDH3 observed in vivo was confirmed in GBM patients from various independent cohorts. In summary, this work describes the functional and mechanistic roles of CDH3/P-cadherin in GBM, establishing it as a negative prognostic biomarker. Future studies are warranted to understand the mechanisms underlying CDH3/P-cadherin implication in tumor aggressiveness and to explore the therapeutic value of novel treatments targeting P-cadherin in GBM.Os gliomas incluem diferentes tumores cerebrais, sendo o glioblastoma (GBM) a forma mais comum e maligna em adultos. O tratamento inclui remoção cirúrgica e radioquimioterapia com o agente alquilante temozolomida. No entanto, os pacientes apresentam um mau prognóstico com uma mediana de sobrevida de 15 meses. A elevada heterogeneidade dos GBMs contribui parcialmente para a ineficácia da terapia atualmente utilizada, pelo que os GBMs são ainda considerados incuráveis. A identificação de novos biomarcadores poderá ajudar num prognóstico mais preciso e na estratificação de pacientes, permitindo o seu direcionamento para métodos terapêuticos mais personalizados. As caderinas são moléculas de adesão celular dependentes de cálcio, cuja implicação em cancro tem sido largamente estudada nas últimas duas décadas. A P(Placental)-caderina, codificada pelo gene CDH3, é uma caderina clássica, que diversos autores têm reportado como promotora ou supressora tumoral, de acordo com o tipo de cancro e com o contexto molecular. Este estudo teve como objetivo compreender as implicações funcionais, mecanísticas e prognósticas de CDH3/P-caderina em gliomas e, mais especificamente, em GBM. O gene CDH3 e a proteína P-caderina estão sobre-expressos num conjunto de gliomas de elevado grau, sugerindo que a P-caderina pode desempenhar funções que aumentam a agressividade dos gliomas. Assim, avaliámos o impacto funcional de CDH3/P-caderina em GBM in vitro. Verificámos que a P-caderina regula diferentes características-chave do cancro em GBM, estando associada a um aumento da viabilidade celular e das propriedades estaminais. Adicionalmente, a P-caderina demonstrou ter um impacto na capacidade de invasão celular. O comportamento oncogénico observado in vitro foi validado in vivo num modelo ortotópico de ratinho de GBM, observando-se que animais injetados com células de GBM a sobre-expressar CDH3 mostraram uma menor sobrevida. Análises mecanísticas demonstraram que os genes positivamente correlacionados com CDH3 em GBM estão enriquecidos para vias de sinalização associadas a cancro. Por fim, é importante salientar que o valor prognóstico de CDH3 observado in vivo foi validado em pacientes com GBM de diferentes coortes. Em suma, este trabalho descreve as implicações funcionais e mecanísticas de CDH3/P-caderina em GBM, estabelecendo-a como um biomarcador de prognóstico negativo. Estudos adicionais são necessários para avaliar os mecanismos pelos quais a P-caderina está associada à agressividade tumoral e para explorar o valor terapêutico da inibição da P-caderina em GBM.The work presented in this thesis was performed in the Life and Health Sciences Research Institute (ICVS), Minho University.
