Searching for potencial antibiotic agents of natural and synthetic origin and their interaction with amyloid fibril formation systemsBacillusCarbohydrate based antibioticsNeuroprotectorsS. sclareoidesAmyloid fibrilsRosmarinic acidTeses de mestrado - 2012Tese de mestrado em Química (Química, Saúde e Nutrição), apresentada à Universidade de Lisboa, através da Faculdade de Ciências, 2012New molecules from synthetic and natural origin, capable of dealing with conditions concerning ageing and/or infection, were investigated by the following approaches: Synthesis of non-toxic alkyl 2,6-deoxy glycosides with potential antibiotic and anti-amyloidogenic activity Identification of active principles of S. sclareoides regarding its anti-amyloidogenic effects.Infections are frequently more severe in elderly population and antibiotics endorsing active and healthier ageing become relevant. Antimicrobial activity was assessed by microdilution method, and the anti-amyloidogenic activity was assessed by fibrilisation/defibrilisation of cystatin B, monitored with ThT and fluorescence spectroscopy. Moreover, plant extracts, their pure components and synthetic glycosides were added to 15N cystatin B and HSQC spectra showed binding to the protein in loop residues and β strands. The ability of synthetic compounds, extracts and pure components to prevent conversion of PrPC to PrPSc was measured by dot blot analysis.
