Contudo o seu papel in vivo nas bactérias onde ocorrem não foi ainda esclarecido. Em Desulfovibrio Vulgaris Hildenborough existem duas Hcp’s, Hcp1 e a Hcp2, que apresentam elevada homologia da sequencia proteica e cuja função e ainda desconhecida. Neste trabalho estudou‐se o papel das Hcp’s de D. vulgaris Hildenborough na resposta ao stress causado pelo oxigénio atmosférico, pelo peroxido de hidrogénio e pelo oxido nítrico. No stress com oxigénio e com peroxido não se observaram diferenças na viabilidade dos mutantes para os genes hcp1 e hcp2 em relação a estirpe selvagem. Os ensaios de complementação em Escherichia coli LMS2710 (estirpe mutada numa redutase de nitrogénio) realizados com os genes hcp1 e hcp2 indicam também que as duas proteínas não contribuem para a resistência de D. vulgaris Hildenborough a este stress. Assim os dados obtidos mostram que estes genes não tem função nestas condições. Averiguou‐se o papel dos genes roo1 e roo2 de D. vulgaris Hildenborough que codificam duas proteínas flavodiiron que estão propostas estar envolvidas na destoxificação do óxido nítrico. Verificou‐se que a expressão da Roo1 e capaz de complementar a mutação da estirpe de Escherichia coli referida (deletada na redutase de nitrogénio). Contudo as estirpes de Desulfovibrio vulgaris Hildenborough mutadas em hcp1 e hcp2, quando submetidas a stress pelo NO, não mostraram diferenças de viabilidade quando comparadas com a estirpe selvagem nas mesmas condições. Outro objetivo deste trabalho foi avaliar a afinidade do regulador HcpR para as zonas promotoras de hcp1 e hcp2. Para isso procedeu‐se a clonagem do gene hcpR de D.vulgaris Hildenborough em pET28a e a expressão e purificação da proteína recombinante. Realizaram‐se ensaios de Electrophoretic Mobility Shift Assay não só com estas zonas promotoras, mas também zonas promotoras dos genes flavodoxina (fld), ferrous iron transporter (feoA) e alkyl hydroperoxide reductase C (ahpC) de Desulfovibrio vulgaris Hildenborough que são descritos como sendo regulados pela HcpR. Embora se tenha observado a alteração da mobilidade das sondas hcp1, hcp2 e feoA na presença de HcpR, apenas para hcp2 foi possível demonstrar que a mobilidade varia com o aumento da quantidade de proteína. Foi assim possível provar a ligação do regulador HcpR a zona promotora do gene hcp2.The Hybrid Cluster Proteins are iron‐sulfur proteins that have very specific properties as having a unique cluster (hybrid cluster) and are described as having hydroxylamine reductase activity. However its role in vivo in the bacteria where they occour as not yet been clarified. In Desulfovibrio vulgaris Hildenborough there are two Hcp's, Hcp1 and Hcp2, which show high protein sequence homology and whose function is still unknown. In this work, we studied the role of the Hcp’s from D.vulgaris Hildenborough’s in response to stress caused by atmospheric oxygen, hydrogen peroxide and nitric oxide. In the stress with oxygen and peroxide no differences were observed in the viability of the hcp1 and hcp2 gene mutants when compared to the wild-type. Complementation assays In Escherichia coli LMS2710 (strain mutated in a nitrogen reductase) performed with the hcp1 and hcp2 genes also indicate that the two proteins do not contribute to the resistance of D.vulgaris Hildenborough to this stress. Thus the data obtained show that these genes do not function under these conditions. The role of the roo1 and roo2 genes of D. vulgaris Hildenborough, flavodiiron encoding two proteins which are proposed to be involved in detoxification of nitric oxide, was examined. It was found that the expression of Roo1 is capable of complementing the mutation of the strain of Escherichia coli above mentioned (deletion in nitrogen reductase ). However strains of Desulfovibrio vulgaris Hildenborough mutated in the hcp1 and hcp2 genes, when subjected to stress by NO, showed no differences in viability when compared with the wild type strain under the same conditions. Another objective of this study was to evaluate the affinity of the HcpR regulator for the promotor regions of hcp1 and hcp2. For this we proceeded to the cloning of the gene hcpR from D.vulgaris Hildenborough in pET28a and its’s expression with the follow purification of the recombinant protein. Electrophoretic Mobility Shift Assays were performed not only with these promotor regions, but also with the promoter regions of the genes flavodoxin (fld), ferrous iron transporter A sub‐unit (feoA) and alkyl hydroperoxide reductase C (ahpC) from Desulfovibrio vulgaris Hildenborough which are described as being regulated by HcpR. We observed the change of the hcp1, hcp2 and feoA probe’s mobility in the presence of HcpR, but only for hcp2 was possible to demonstrate that the mobility varies with the increased amount of protein.
