Resumo: | Este estudo teve como objectivo caracterizar as alterações do proteoma nuclear da linha celular HC11, um modelo utilizado no estudo do cancro da mama, durante o seu processo de diferenciação. No sentido de avaliar a variação do perfil de modificações pós-traducionais em histonas com o processo de diferenciação, procedeu-se à purificação de núcleos e isolamento de histonas de células HC11 stem-cell like e diferenciadas. O perfil de modificações pós-traducionais em histonas foi analisado com recurso a HPLC-ESI-MS/MS e nanoLC-MALDIMS/MS, após digestão com tripsina e Glu-C, tendo-se identificado um total 36 variantes de histonas e 2659 modificações, 99% das quais com um espectrómetro de massa do tipo trapa linear com fonte de electrospray. A diminuição do grau de acetilação nas classes de histonas H3 e H4, detectada por AUT-PAGE, a diminuição do número de resíduos de aminoácidos acetilados e o aumento dos metilados em H3 e H4, a presença do resíduo trimetilado H3K27 apenas nas células diferenciadas, a identificação da H3K9 acetilada e de H3K4 dimetilada apenas nas células estaminais suportam a noção de que o grau de diferenciação celular traduz-se por uma diminuição da capacidade de expressão génica. No entanto, o número mais elevado de PTMs diferentes observado no N-terminal da H4 sugere uma importante, e ainda desconhecida, contribuição desta classe de histonas na regulação epigenética. O proteoma nuclear foi analisado por electroforese bidimensional e identificaram-se, usando um MALDI-TOF/TOF, 100 proteínas distintas, 48% das quais referentes à classe da “biossíntese de DNA/RNA/Proteínas”. De entre as 5 proteínas diferentemente expressas entre as células estaminais e diferenciadas, é de salientar a maior expressão nas células diferenciadas das hnRNPs associadas ao processo de apoptose e proliferação celular. A identificação de diferenças de expressão de proteínas relacionadas com o cancro, em conjunto com modificações pós-traducionais identificadas, nomeadamente a presença de H3K9ac apenas nas células estaminais, corrobora o uso das células HC11 como modelo para o estudo do cancro da mama. ABSTRACT: The aim of the present work was to characterize the changes in the nuclear proteome of the cell line HC11, a model used in the study of breast cancer during their differentiation process. To assess the alterations in the post-translational modifications’ profile in histones with the differentiation process, we performed the purification of nuclei and isolation of histones from HC11 stem-cell like and differentiated cells. The profile of post-translational modifications in histones was analyzed by HPLC-ESIMS/ MS and nanoLC-MALDI-MS/MS, after digestion with trypsin and Glu-C, being identified a total of 36 variants of histones and 2659 PTMs, 99% of which with a linear ion trap mass spectrometer with an electrospray source. The decrease in the degree of histone acetylation in H3 and H4 classes, analysed by AUT-PAGE, the diminished number of amino acid residues, the increase of acetylated H3 and methylated H4, the presence of trimethylated H3K27 in differentiated cells, the identification of acetylated H3K9 and dimethylated H3K4 in stem cell like support the notion that cellular differentiation is reflected by a decreased ability to gene expression. However, the highest number of PTMs observed in the N-terminal of H4 from differentiated cells suggests an important, yet unknown, contribution of this class of histone in epigenetic regulation. The nuclear proteome was analyzed by twodimensional electrophoresis and 100 distinct proteins were identified using a MALDI-TOF/TOF, 48% of which were related with “biosynthesis of DNA/RNA/Protein”. Among the five proteins differentially expressed between stem-cell like and differentiated cells, it is worth of note the higher levels of hnRNPs associated with apoptosis and cell proliferation, in differentiated cells. The identification of expression differences in proteins related with cancer, together with the post-translational modifications identified, which includes the presence of H3K9ac in stem cell like, supports the use of HC11 cells as a model for the study of breast cancer. |
