Genetic analysis of Schistosoma mansoni sensitive and resistant strains to praziquantel using rapd-pcr

Resumo: Praziquantel (PZQ) is the drug of choice in the treatment of schistosomiasis due to its high cure rates and no significant side effects. Currently, more and more cases of resistance or increased tolerance to PZQ have been reported, consequently, there were recommendations for continuous monitoring of drug resistance. In our laboratory, we selected a parasitic line of Schistosoma mansoni, through constant drug pressure over several cycles, which was resistant to 120 mg/kg PZQ. Adult parasites of S. mansoni from the resistant and from a sensible strain were collected; the DNA was extracted and analyzed through Random Amplified Polymorphic DNAPolymerase Chain Reaction (RAPD-PCR). We observed genetic polymorphism in 7 of 10 primers tested. Once observed the polymorphic differences between susceptible and resistant parasites, we calculated the similarity coefficient (Dice’s coefficient). Understanding the genetic polymorphism associated with resistance may contribute to the future identification of related sequences with the resistance/tolerance of S. mansoni to PZQ treatment and for identification of resistant parasites in the field as well as for the development of new strategies to schistosomiasis control.

Devido a estas observações, a OMS recomenda a monitorização contínua de Schistosoma spp potencialmente resistentes ao fármaco. No laboratório de Helmintologia foi seleccionada uma linha de Schistosoma mansoni, resistente a PZQ na dose de 120 mg/Kg, após pressão de fármaco constante ao longo de vários ciclos. Foram recolhidos parasitas adultos de S. mansoni da estirpe resistente e de uma estirpe sensível e procedeu-se à análise do DNA dos vermes por Random Amplified Polymorphic DNA-Polymerase Chain Reaction (RAPD-PCR). Foi possível observar polimorfismos em 7 dos 10 primers. Uma vez observadas as diferenças polimórficas entre parasitas sensíveis e resistentes, calculou-se o coeficiente de similaridade (Dice’s coefficient).
