Impacto da rinite alérgica na atividade da sinaptofisina na via olfativa

Resumo: ABSTRACT Introduction: Allergic rhinitis (AR) has been pointed out as a cause of olfactory dysfunction. In addition to the classic symptoms, AR has also been associated with alterations in the normal sleep pattern, decreased cognitive performance and increased likelihood of depression and anxiety. Although the mechanisms underlying these associations remain undetermined, the olfactory pathway has been proposed as a possible link between chronic nasal inflammation and the occurrence of changes in the central nervous system. Thus, this study aims to investigate the influence of AR on synaptophysin activity in the olfactory pathway and prefrontal cortex, by immunohistochemistry, in an animal model of AR. We also aimed to evaluate the influence of AR on olfactory function and aggressive behaviors in mice with AR. Methods: An AR model was replicated in male Wistar rats by sensitization with ovalbumin. After olfactory and behavioral tests, we studied the synaptic structure of the olfactory bulb, anterior olfactory nucleus, piriform cortex, and prefrontal cortex, by detecting synaptophysin (Syn) labeling by immunofluorescence. Results: There was a significant decrease in Syn labeling in the glomerular layer of the olfactory bulb and in the prefrontal cortex, in rats with AR. Moreover, olfactory evaluation revealed significant olfactory dysfunction in the AR group. Behavioral tests showed a predominance of defensive behaviors in AR rats. Conclusions: In this study we found that AR influences the activity of Syn in the olfactory pathway and CPF, leading to decreased olfactory function and altering the aggressive behavior of rats with AR.
