These functions make it a versatile tool that may be applied in various scientific fields. In recent years, these molecules have arisen a special interest in the scientific community due to their potential in the development of innovative therapies in vivo, in particular in immunotherapy. Regarding solid tumours, their therapeutic application is extremely interesting once they make immunomodulation possible directly in the unregulated tumour microenvironment.This work explores the particular characteristics of mRNA molecules and the different factors that can influence their effectiveness, especially in oncology. To that end, it is not only necessary to guarantee its safety as well as quality during its manufacturing process, but also is imperative to promote an effective delivery to the action site. The chosen delivery system must be able to overcome all biological barriers, avoid its extra and intracellular degradation and hence bring the therapeutic agent to the target in therapeutically relevant concentrations. In fact, efficient delivery remains a technical barrier to the application of mRNA molecules in vivo, only being somewhat overtaken by some non-viral drug delivery vehicles such as lipid nanoparticles, charge-altering releasable transporters and other few systems. Although gene therapy has been used for over 30 years, nucleic acids have only recently been applied therapeutically in humans. Antibodies, proteins and cells can be considered the therapeutic agents of choice in immunotherapy in cancer treatment. In fact, all the advantages they bring to clinical practice are unquestionable, however, immunotherapy still has room for improvement regarding their safety and efficacy.As estruturas bioquímicas à base de RNA mensageiro desempenham diferentes funções no nosso organismo, desde a regulação da expressão genética à capacidade de desencadearem efeitos pró-inflamatórios. São, portanto, uma ferramenta útil, inovadora e versátil com uma vasta aplicação. Recentemente, a ciência reconheceu nestas moléculas um enorme interesse, devido ao potencial que apresentam no desenvolvimento de terapêuticas inovadoras in vivo, em particular na imunoterapia. Relativamente aos tumores sólidos, a aplicação destas moléculas na terapêutica é especialmente interessante uma vez que estas permitem induzir uma ação imunomoduladora diretamente no microambiente tumoral desregulado.As características particulares destas moléculas e os fatores que influenciam a sua eficácia, serão discutidas neste trabalho, incidindo especialmente na área da oncologia. Por forma a garantir tratamentos eficazes, não só é fundamental garantir a segurança e qualidade destas moléculas durante o processo de fabrico, mas também tornar possível a sua entrega no local de ação. O veículo de entrega escolhido deverá ser capaz de transpor barreiras biológicas, evitar a sua degradação tanto extra como intracelular para assegurar que o agente terapêutico atinge o local de ação em concentrações terapeuticamente relevantes.
