A primeira parte deste documento consiste no relatório de estágio relativo ao estágio que realizei em farmácia comunitária, nomeadamente na farmácia Hebel. Este relatório é apresentado sob a forma de uma análise SWOT que identifica os pontos fortes, os pontos fracos, as oportunidades e as ameaças inerentes à realização deste estágio. Para além disso, nesta parte é ainda apresentado cinco casos práticos que espelham situações que surgiram no contexto de atendimento, nas quais tive a possibilidade de aplicar os conhecimentos adquiridos até então. Da mesma forma, numa segunda parte consta o relatório de estágio referente ao estágio que realizei em indústria farmacêutica, designadamente, na Bluepharma®, apresentado também sob a forma de uma análise SWOT. Por último, a terceira parte compreende a monografia por mim elaborada, intitulada “Blood-brain barrier-on-a-chip for central nervous system disease modeling and drug screening”. Efetivamente, a barreira hematoencefálica é uma estrutura complexa e dinâmica que regula as moléculas que passam do sangue para o sistema nervoso central, mantendo a homeostase cerebral. No entanto, esta também constitui um obstáculo à entrega de fármacos. Adicionalmente, a disrupção da barreira hematoencefálica está associada a várias doenças neurológicas. Assim, modelos in vitro da barreira hematoencefálica que permitam o estudo da função e disfunção desta barreira são essenciais para investigação na área biomédica e no desenvolvimento de fármacos que tenham como alvo o sistema nervoso central. Nesse sentido, BBB-on-chips são modelos in vitro promissores que replicam mais fisiologicamente o ambiente in vivo da barreira hematoencefálica, permitindo obter resultados mais representativos e transponíveis para a situação clínica. Assim, nesta monografia, são descritos os diferentes componentes dos BBB-on-chips e as estratégias que têm sido utilizadas para os otimizar ao longo dos últimos anos.This document was elaborated within the scope of the curricular unit “Curricular Internship”, included in the study plan of the Integrated Master’s in Pharmaceutical Sciences from the Faculty of Pharmacy, and it is divided into three parts. The first part of this document consists of the internship report of the internship I carried out in community pharmacy, namely, in the Hebel pharmacy. This report is presented in the form of a SWOT analysis which identifies the strengths, weaknesses, opportunities, and threats inherent to the completion of this internship. Furthermore, in this part, I have also presented five practical cases that reflect situations that occurred in the attendance context, in which I had the opportunity to apply the knowledge acquired so far. In the same way, a second part contains the internship report referring to the internship I carried out in pharmaceutical industry, specifically in Bluepharma®, also presented in the form of a SWOT analysis. Finally, the third part comprises my monograph entitled “Blood-brain barrier-on-a-chip for central nervous system disease modeling and drug screening”. Effectively, the blood-brain barrier (BBB) is a complex dynamic structure that regulates the molecules that pass from the blood to the central nervous system (CNS), maintaining brain homeostasis. However, it also constitutes an obstacle to drug delivery. In addition, BBB disruption is associated with various neurologic diseases. Thus, in vitro BBB models that allow the study of BBB function and dysfunction are essential for biomedical research and the development of drugs targeting the CNS. In that regard, BBB-on-chips are promising in vitro models that replicate the in vivo BBB microenvironment more physiologically, allowing more representative and transferable results to the clinical situation.
