BIT by BIT: Associating Transcription to Ageing

Resumo: | A common and longstanding view of the ageing process poses that the onset of ageing hallmarks is driven by the gain of somatic mutations deriving from the inaccurate repair of DNA lesions, the most catastrophic of which is Double-strand Break (DSB). However, compelling evidence accumulated during the last decade demonstrated that, in addition to causing mutations, DSBs can initiate gene expression events. Using live-cell imaging, it was recently provided direct evidence of transcription initiation at DSB sites, a process dubbed Break-Induced Transcription (BIT), which was previously reported as an important step of DSB repair and gene expression regulation. A trademark of the ageing cell is the widespread rewiring of gene expression, including an increase in spurious transcription. This led us to propose the disruptive hypothesis that alternative gene expression events established through BIT at DSB contribute to the establishment of the ageing phenotype. By using public human data from genome-wide detection techniques of DSB (BLISS, DSBCapture, Break-seq) and high-throughput sequencing (RNA-seq), whose cells under- went no treatment, it was possible to identify natural BIT events at the whole genome level. We were able to develop a genome-wide detection tool of BIT independently of the DSB technique resolution. The available dataset provided no evidence of BIT events influencing ageing itself. |

The available dataset provided no evidence of BIT events influencing ageing itself.Uma visão comum e duradoura do processo de envelhecimento sugere que o início das marcas de envelhecimento é uma consequência pela acumulação de mutações somáticas resultantes da reparação imprecisa de lesões no DNA, sendo a mais catastrófica delas a quebra de dupla hélice (DSB). No entanto, evidências convincentes acumuladas durante a última década demonstraram que, além de causar mutações, as DSBs podem levar a eventos de expressão genética. Através de microscopia em tempo real de células vivas, foi recentemente obtido provas da iniciação da transcrição nos locais de DSB, um processo denominado Indução de Transcrição por Quebra (BIT), que anteriormente foi relatado como uma etapa importante na reparação de DSB e na regulação da expressão genética. Uma característica da célula envelhecida é a reconfiguração generalizada da expressão genética, incluindo um aumento na transcrição espúria. Isso levou-nos propor a hipótese disruptiva de que eventos alternativos de expressão genética através de BIT em DSB contribuem para o estabelecimento do fenótipo de envelhecimento. Através do uso de dados públicos de humanos de técnicas de detecção de DSB em todo o genoma (BLISS, DSBCapture, Break-seq) e sequenciação de RNA de nova geração (RNA-seq), cujas células não foram submetidas a qualquer tratamento tratamento, foi possível identificar eventos naturais de BIT em todo o genoma. Fomos capazes de desenvolver uma ferramenta de detecção de BIT em todo o genoma, independentemente da resolução da técnica de DSB.
