Although COVID-19 vaccination is being effective, the emergence of new variants of concern has been compromising vaccine efficacy. Therefore, there is an urgent need to develop new therapeutic options and to better understand the virus transmission and their reservoirs. Recent studies demonstrated that domestic cats are susceptible to SARS-CoV-2 infection and can be naturally and experimentally infected with SARS-CoV-2. In this context, this study aimed to understand and characterise the humoral response of domestic cats naturally infected with SARS-CoV-2 and to evaluate their suitability to develop neutralizing antibodies against the virus. For this purpose, serum samples from 614 cats were collected during medical appointments at the School Hospital of Faculty of Veterinary Medicine - University of Lisbon between May and October 2021. These serum samples were analysed for the presence of IgG antibodies against SARS-CoV-2 by evaluating antibodies that specifically bind to the RBD and spike viral proteins. From the 614 cats analysed, 15.8% (97/614) tested positive for the presence of IgG antibodies to SARS-CoV-2 RBD and spike proteins. An in-vitro characterization of the positive samples was followed by the detection of neutralizing antibodies to wild type and delta variants by using a serum neutralisation assay. Of the SARS-CoV-2 seropositive animals, 21.6% (21/97) were positive on serum neutralisation assay. Later, serial dilutions of these 21 serum samples were performed for the determination of the antibody titre. Furthermore, the 21 serum samples were then tested in ELISA and neutralisation assays for the new variant of concern, omicron. The 21 serum samples had antibodies that bind to the RBD of omicron variant, but only 4 of this samples had neutralising antibodies to this variant. As reported in several recent studies, we also identified a potential SARS-CoV-2 human-to-cat transmission and detected anti-SARS-CoV-2 antibodies with neutralising properties in cat serum samples. The results obtained in this study suggest that the antibodies with the best characteristics may become useful in mapping the binding epitopes of the spike protein and, therefore, may constitute a promising approach for the development of new therapeutic molecules for COVID-19.RESUMO - Caracterização da resposta humoral de gatos domésticos naturalmente infetados com SARS-CoV-2 - Em 2019, surgiu um novo coronavírus, o coronavírus da síndrome respiratória aguda grave 2 (SARS-CoV-2), responsável pela pandemia da doença do coronavírus 2019 (COVID-19). Embora a vacinação esteja a ser uma medida eficaz, o aparecimento de novas variantes de preocupação tem vindo a comprometer a eficácia da vacina. Por isso, há uma necessidade de desenvolver novas opções terapêuticas e de compreender melhor a transmissão e os hospedeiros deste vírus. Estudos recentes demonstraram que os gatos domésticos são suscetíveis à infeção por SARS-CoV-2, podendo ser natural ou experimentalmente infetados com SARS-CoV-2. Neste contexto, este estudo tem como objetivo perceber e caracterizar a resposta humoral de gatos domésticos naturalmente infetados com SARS-CoV-2 e avaliar a sua capacidade de desenvolver anticorpos neutralizantes contra o vírus. Para isso, foram recolhidas amostras de soro de 614 gatos durante as consultas médicas no Hospital Escolar da Faculdade de Medicina Veterinária – Universidade de Lisboa, entre maio e outubro de 2021. As amostras de soro foram analisadas quanto à presença de anticorpos IgG contra o SARS-CoV-2 através da avaliação dos anticorpos que ligavam especificamente às proteínas virais RBD e spike. Dos 614 gatos analisados, 15.8% (97/614) testaram positivo para a presença de anticorpos IgG para o SARS-CoV-2. De seguida, foi feita uma caracterização in vitro das amostras positivas para deteção de anticorpos neutralizantes para as variantes wild type e delta através de ensaios de neutralização. Dos animais seropositivos para o SARS-CoV-2, 21.6% (21/97) foram positivos no ensaio de neutralização. Depois foram feitas diluições seriadas destas 21 amostras de soro para se determinar o título de anticorpos. Além disso, as 21 amostras foram testadas em ensaios de ELISA e de neutralização para a nova variante de preocupação, ómicron. As 21 amostras apresentavam anticorpos que ligavam à RBD da variante ómicron, mas apenas 4 destas amostras tinham anticorpos neutralizantes. Tal como reportado em diversos estudos recentes, também identificamos uma potencial transmissão humano para gato e detetamos anticorpos anti-SARS-CoV-2 com propriedades neutralizantes nas amostras de soro de gato.
