Cytotoxicity of Electrospun mats loaded with antimicrobial agents towards Macrophages

Resumo: | New developments in nanotechnology have transformed wound treatment by introducing materi- als with better absorption and antimicrobial properties. Biocompatible nanoparticles, like gold (Au), silver (Ag), and zinc (Zn), have become promising candidates for promoting wound healing. This re- search used a chemical reduction method to create Au, Ag, and Zn nanoparticles. Additionally, nano- fibers were made using electrospun solutions consisting of polycaprolactone (PCL), chitosan (CS), and gelatine (GEL), which included varying amounts of nanoparticles and antiseptics like povidone-iodine (PVP-I), chlorhexidine (CHX), and octenidine dihydrochloride (OCT). The study examined these na- noparticles, polymer-based materials, and antiseptics in detail, highlighting their various interactions. The findings have significant implications for biomedical science, antimicrobial applications, and the development of biomaterials. Among the nanoparticles studied, AuNPs stood out as excellent candidates for applications that require precise particle sizes with remarkable uniformity. ZnONPs also showed size uniformity, while AgNPs were slightly less consistent but still within acceptable ranges. Further analysis using scanning electron microscopy (SEM) and dynamic light scattering (DLS) revealed how various additives affected fibre morphology within PCL and GEL mats. Some additives led to better fibres, while others created broader distributions of fibre diameters. In chitosan (CS) mats, cross-linking reduced fibre diameter without compromising mat structure, and additives had minimal impact on fibre morphology. Cytotoxicity tests showed that some formulations were non-cytotoxic while others had varying degrees of cytotoxicity. Finally, the antibacterial properties of AgNPs, AuNPs, and ZnONPs against E. coli were tested, with AgNPs showing promise in fighting bacterial infections, especially at lower concentrations. |

As nanopartículas biocompatíveis, como o ouro (Au), a prata (Ag) e o zinco (Zn), tornaram-se candidatos promissores para promover a cicatrização de feridas. Esta investigação utilizou um método de redução química para criar nanopartículas de Au, Ag e Zn. Além disso, foram produzidas nanofibras utilizando soluções electrfiadas constituídas por policaprolactona (PCL), quitosano (CS) e gelatina (GEL), que incluíam quantidades variáveis de nanopartículas e anti-sépticos como iodopovidona (PVP-I), clorexidina (CHX) e dicloridrato de octenidina (OCT). O estudo examinou em pormenor estas na-nopartículas, materiais à base de polímeros e anti-sépticos, destacando as suas várias interacções. Os resultados têm implicações significativas para a ciência biomédica, as aplicações antimicrobianas e o desenvolvimento de biomateriais. Entre as nanopartículas estudadas, as AuNPs destacaram-se como excelentes candidatos para aplicações que requerem tamanhos de partículas precisos com uma uniformidade notável. As ZnONPs também mostraram uniformidade de tamanho, enquanto as AgNPs foram um pouco menos consistentes, mas ainda dentro de faixas aceitáveis. Análises posteriores utilizando microscopia eletrónica de varrimento (SEM) e dispersão dinâmica da luz (DLS) revelaram como os vários aditivos afectaram a morfologia das fibras nos tapetes de PCL e GEL. Alguns aditivos conduziram a fibras mais finas, enquanto outros criaram distribuições mais alargadas de diâmetros de fibras. No que diz respeito às fibras do quitosano (CS), a reticulação reduziu o diâmetro das fibras sem comprometer a estrutura da membrana, e os aditivos tiveram um impacto mínimo na morfologia das fibras. Os testes de citotoxicidade mostraram que algumas formulações não eram citotóxicas, enquanto outras apresentavam graus variáveis de citotoxicidade.
