Resumo: Experimental evidence suggests the participation of the cerebellum in motor and non-motor functions. Since recent results demonstrate the participation of the histaminergic cerebellar system in the consolidation of memory, this study investigated the effect and the understanding of the molecular bases of the histaminergic H4 agonist (VUF-8430) microinjection in the consolidation of emotional memory in mice submitted to the elevated plus maze (EPM) and the inhibitory avoidance task (IAT) as well as in motor control in mice submitted to the rotarod test. On the fourth day after surgical recovery from the procedure to implant a cannula, the animals received saline (SAL) or VUF (0.15 nmol; 0.49 nmol; e 1.48 nmol/0.1 μl) administered post-exposure. Experiments 1, 2 and 3 were performed on two consecutive days: Exposure and 24h after, the re-exposure. Experiment 1: was held in the EPM and immediately post-T1, the pharmacologic treatment occurred; in T2, there were only re-exposure to EPM. The decrease in activity in the open arms (OAE and OAT) between T1 and T2, inferred the emotional memory. Experiment 2: was held in the IAT and the pharmacological treatment was post-D1; in D2, the animals were only re-exposed to the IAT. The increase or decrease of latencies in D2 compared to D1 inferred the emotional memory. Experiment 3: was held in the rotarod and the animals received the drug treatment post-R1; in R2, the animals were only re-exposed, featuring motor learning. The increase or decrease in R2 latencies compared to R1, inferred the motor learning. Experiment 4: VUF-8430 was I.P. injected at a dose of 500 ng/kg to investigate protein expression by phosphorylation of the CREB transcription factor, using the Western blotting technique, in the cerebellar vermis and in classic structures linked to emotional memory (prefrontal cortex, amygdala and hippocampus). Results: in Experiment 1, VUF (0.49 nmol and 1.48 nmol) impaired the consolidation of emotional memory in the EPM; in Experiment 2, VUF (1.48 nmol) impaired the consolidation of emotional memory in the IAT; in Experiment 3, VUF (0.49 nmol and 1.48 nmol) impaired the consolidation of motor learning in the rotarod; in Experiment 4, there was a decrease in protein expression (CREB and pCREB) in the cerebellar vermis and prefrontal cortex in mice. Conclusions: the highest dose of VUF-8430 (1.48 nmol), microinjected in the cerebellar vermis in mice, induced deficit in learning and memory consolidation in the models used in the present study (EPM, IAT and rotarod). In addition, the decrease in the protein expression in the cerebellar vermis verified by CREB and pCREB levels when injected I.P. the highest dose of the H4 receptor agonist (VUF-8430) can corroborate these deficits.

Visto que recentes resultados demonstram a participação do sistema cerebelar histaminérgico na consolidação da memória, este trabalho investigou o efeito de microinjeções e o entendimento das bases moleculares do agonista histaminérgico H4 (VUF-8430) na consolidação da memória emocional em camundongos submetidos ao labirinto em cruz elevado (LCE), um modelo voltado à ansiedade (Experimento 1); à caixa de esquiva inibitória (EI), um modelo voltado ao medo (Experimento 2); no controle motor e na aprendizagem motora em camundongos submetidos ao rotarod (Experimento 3); e, na expressão proteica dos níveis séricos de CREB e CREB fosforilado através da técnica de Western blotting (Experimento 4). No quarto dia após a cirurgia de implantação da cânula-guia, para a realização dos Experimentos 1, 2 e 3, os animais receberam microinjeções intra-vérmis de salina ou VUF (0,15, 0,49 e 1,48 nmol/0,1 μl) pós-exposição. Esses experimentos foram realizados em dois dias consecutivos. Experimento 1: foi realizado no LCE e, imediatamente após a exposição (T1), houve o tratamento farmacológico; no segundo dia de teste (T2), ocorreu apenas a reexposição ao LCE. A diminuição da atividade nos braços abertos (EBA e TBA) em T2 comparada à T1, permitiu inferir a aprendizagem e consolidação da memória emocional. Experimento 2: foi realizado na EI e o tratamento farmacológico ocorreu após o primeiro dia de teste (D1); no segundo dia de teste (D2), os animais foram apenas reexpostos à EI. O aumento ou a diminuição das latências de D2 comparado à D1, permitiu inferir a facilitação ou o déficit na aprendizagem e consolidação da memória emocional. Experimento 3: foi realizado no rotarod e os animais receberam o tratamento farmacológico imediatamente após a exposição (R1); no segundo dia de teste (R2), os animais foram apenas reexpostos ao aparato, caracterizando a aprendizagem motora. O aumento ou a diminuição das latências de R2 comparado à R1 permitiu inferir a facilitação ou déficit na consolidação da aprendizagem motora. Experimento 4: foi realizada a injeção sistêmica intraperitoneal (I.P.) do VUF-8430, na dose de 500 ng/kg, para investigar a expressão proteica através da fosforilação do fator de transcrição CREB, utilizando a técnica de Western blotting, no vérmis cerebelar e em estruturas clássicas vinculadas à memória emocional (córtex pré-frontal, amídala e hipocampo). Resultados: no Experimento 1, o VUF (0,49 nmol e 1,48 nmol) prejudicou a consolidação da memória emocional no LCE; no Experimento 2, o VUF (1,48 nmol) prejudicou a consolidação da memória emocional na EI; no Experimento 3, o VUF (0,49 nmol e 1,48 nmol) prejudicou a consolidação da aprendizagem motora no rotarod; e, no Experimento 4, houve uma diminuição da expressão proteica (CREB e pCREB) no vérmis cerebelar e no córtex pré-frontal de camundongos. Conclusões: a maior dose do VUF-8430 (1,48 nmol), microinjetada no vérmis cerebelar de camundongos, induziu um déficit na aprendizagem e consolidação da memória nos modelos utilizados no presente estudo. Experiment 4: VUF-8430 was I.P. injected at a dose of 500 ng/kg to investigate protein expression by phosphorylation of the CREB transcription factor, using the Western blotting technique, in the cerebellar vermis and in classic structures linked to emotional memory (prefrontal cortex, amygdala and hippocampus). Results: in Experiment 1, VUF (0.49 nmol and 1.48 nmol) impaired the consolidation of emotional memory in the EPM; in Experiment 2, VUF (1.48 nmol) impaired the consolidation of emotional memory in the IAT; in Experiment 3, VUF (0.49 nmol and 1.48 nmol) impaired the consolidation of motor learning in the rotarod; in Experiment 4, there was a decrease in protein expression (CREB and pCREB) in the cerebellar vermis and prefrontal cortex in mice. Conclusions: the highest dose of VUF-8430 (1.48 nmol), microinjected in the cerebellar vermis in mice, induced deficit in learning and memory consolidation in the models used in the present study (EPM, IAT and rotarod). In addition, the decrease in the protein expression in the cerebellar vermis verified by CREB and pCREB levels when injected I.P. the highest dose of the H4 receptor agonist (VUF-8430) can corroborate these deficits.
