MICE VIRULENCE ASSAY OF Pasteurella muttocida STRAINS ISOLATED FROM PIGS

Resumo: Virulence of 24 strains of Pasteurella multocida, 13 isolated from pleuritic and pneumonic pig lungs, and 11 from pig lungs obtained from abattoirs was studied. P multocida was cultured in BHI medium incubated at 37° C for 18 h and the number of colony forming units (C FU/ml) determined by inoculation of decimal dilutions on blood agar plates. Dilutions from 10 4 to 10.13 (8,3 x 10 15 to 1 CFU) were inoculated intraperitoneally in mice and the number of deaths for each dilution determined over a 7-day period. Each inoculated strain was recovered from at least two mice. LD50 were determined by the Spearman- Kãrber method. It was observed a wide variation in LD50, ranging from <1 to 10 10.2 CFU. There was no difference in virulence among strains from clinical cases or from abattoir lungs.

Foi analisada a virulência de 13 amostras de Pasteurella multocida isoladas de pulmões de suínos com pneumonia e pleurite e 11 de pulmões de suínos provenientes de frigoríficos. As bactérias foram cultivadas em meio 13H1 por 18 horas a 37° C e realizada a titulação (UFC/ ml) por semeadura de diluições decimais em ágar sangue. Diluições a partir de 10.5 até I (contendo de 1 a 8,3 x 10' 5 UFC) foram inoculadas por via intraperitoneal em camundongos e observado o número de mortes em cada diluição por um período de sete dias. Cada amostra inoculada foi isolada de pelo menos dois camundongos. A DL50 foi calculada pelo método de Spearmann Kãrber e determinado o número de unidades formadoras de colônia (UFC) capaz de matar 50% dos camundongos (D1.50). Ocorreu grande diferença entre as amostras estudadas quanto às DL50, com títulos entre < 1 até 10 102 UFC. Não foi observada diferença de virulência entre as amostras isoladas de casos clínicos e de pulmões de frigoríficos. Departamento de Diagnóstico e Pesquisa Agropecuária.
