PRODUCTION OF ANTISERA FOR DIAGNOSIS OF SOFT-ROTTING PECTOBACTERIAS IN POTATO

Resumo: Polyclonal antisera against Pectobacterium carotovorum subsp. atrosepticum (Pca), P carotovorum subsp. carotovorum (Pcc) and P chrysanthemi (Pch) were produced from intact, untreated cel Is. Bacterial suspensions with 6 to 7 x CFU/ml (2 ml), emulsified with an equal volume of complete Freund's adjuvant were used as antigens to immunize rabbits. The antigens were injected four times at weekly intervala. The antisera obtained (1:640 titer) were specific against the antigens used and did not exhibit any reaction toward other phytopathogenic bacteria, vírus and fungi, as well as potato, tomato, and tobacco plant extracts. Only antigens presents in the cellular structures of Pca, Pcc and Pch are identified by the antibodies and that the latex agglutination test can be used with di I utions of the antigens in order of 1:1 and 1:10 of the initial concentration (50% T, 550 nm).

Foram produzidos anti-soros a partir de células inteiras e não tratadas de Pectobacterium carotovorum subsp. atrosepticum (Pca), P carotovorum subsp. carotovorum (Pcc) e P clzrysanthemi (Pch). Utilizou-se como antígeno . 2 ml de suspensão bacteriana na concentração de 6 a 7 x 10 7 UFC/ml, emulsificada com igual volume de adjuvante completo de Freund, fazendo quatro aplicações em coelhos, com intervalos semanais. Os anti-soros obtidos (título 1:640) foram específicos em relação aos antígenos utilizados e não reagiram com outras bactérias, vírus e fungos fitopatogênicos, bem como, com extratos de plantas de batata, fumo e tomateiro. Concluiu-se que apenas antígenos presentes nas estrutu- ras celulares de Pca, Pcc e Pch são identificados pelos anticorpos produzidos e que o teste de aglutinação em látex pode ser realizado com diluições do antígeno nas proporções de 1:1 e 1:10, partindo-se da concentração padrão (50%T; 550 nm).
