Resumo: | The minerals calcium (Ca) and phosphorus (P) are essential in the diet of laying hens, participating in many metabolic functions, in addition in the diet of laying hens, participating in many metabolic functions, in addition to eggshell formation. However, the different levels can result in a big difference, due to the reduction of the mineral’s absorption capacity. Therefore, the objetictive of this work was to profile the expression of Ca and P-related genes, using duodenum and kidney of older laying hens. Samples were collected from Bovans White laying hens, aged 73 weeks, fron three groups whith different levels of performance and Ca/P retio in the diet, as follows: (HP) high performance (22.43 Ca/P), low performance (HL) 6.71% Ca/P and the standart diet (STD) commomly used 11.43 Ca/P. For the study, RNA sequencing was performed with 27 samples from the duodenum (9/group), using the RNA-Seq technique. For kidney expression, 30 kidney samples (10/group) were selected, evaluation analysis of the chosen genes, being: CYP24A1, FG3R, PTH, TRPV6, CALB1, CALM2, FGF1 and TRPC3. As a result, considering the comparisons between groups, 107 genes were enriched in mineral homeostasis processes, highlighting the genes: CAV1, MYLK, CAB39L, ILIRLI, TNIP3, COL3A1, PKIA, FGF7, ESRRG, PTHLH, ADIPOQ e ESRRG. No rim, os genes CYP24A1 and TRCP3 genes were DE between HP and STD groups and between lp and std, respectively. In this study, we identified new candidate genes for calcium and phosphorus mechanisms in older laying hens. |

In this study, we identified new candidate genes for calcium and phosphorus mechanisms in older laying hens.Os minerais cálcio (Ca) e fósforo (P) são essenciais na dieta de poedeiras, participam de muitas funções metabólicas, além da formação da casca do ovo. Porém, níveis inadequados podem resultar em perdas econômicas significativas após o pico de postura, pela redução da capacidade de absorção mineral. Portanto, o objetivo do trabalho foi analisar o perfil de expressão de genes relacionados com Ca e P, utilizando duodeno e rim de poedeiras com idade avançada. As amostras foram coletadas de poedeiras Bovans White, com 73 semanas, provenientes de três grupos com diferentes níveis de desempenho e relação Ca/P na dieta, sendo: (HP) alto desempenho (22,43 Ca/P), baixo desempenho (HL) 6,71% Ca/P e dieta padrão (SDT) comumente utilizada 11,43 Ca/P. Para o estudo, foi realizado o sequenciamento do mRNA, com 27 amostras do duodeno (9/grupo), utilizando a técnica de RNA-Seq. Para análise de expressão gênica do rim, 30 amostras (10/grupo) foram submetidas a avaliação de oito genes candidatos escolhidos, sendo eles: CYP24A1, FG3R, PTH, TRPV6, CALB1, CALM2, FGF1 E TRPC3. Como resultado, considerando as comparações entre os grupos, 107 genes foram diferencialmente expressos (DE, FDR <0.05) no duodeno. Muitos genes foram enriquecidos em processos de homeostasia mineral, destacando-se os genes: CAV1, MYLK, CAB39L, ILIRLI, TNIP3, COL3A1, PKIA, FGF7, ESRRG, PTHLH, ADIPOQ e ESRRG. No rim, os genes CYP24A1 e TRCP3 foram DE entre os grupos HP e STD e entre LP e STD, respectivamente.
