Resumo: The catingueira (Caesalpinia pyramidalis Tul.) is an endemic species of the Caatinga, which has the potential timber, fodder and medicinal. The plant tissue culture has proved effective in multiplication and conservation of woody species with potential explored. Thus, this study investigated the effects of the types and concentrations of plant growth regulators on in vitro multiplication of C. pyramidalis and develop a protocol for in vitro conservation of the same. For multiplication was carried out three experiments in which we assessed the effects of different concentrations of plant growth regulators (BAP, KIN, TDZ or NAA) on different explants (hypocotyls, cotyledon segments, cotyledons, and epicotyl nodal segments). For the conservation in vitro experiments there were two: firstly tested the effect of different concentrations of sucrose (87.64, 131.46, 175.28 and 219.10mM) supplemented with different concentrations (0.0 and 87.64mM) of sorbitol or mannitol, and second, to evaluate the use of different concentrations (0.0, 1.5, 3.0, 4.5 and 6.0µM) the growth retardant paclobutrazol (PBZ). The culture medium used for all experiments was the WPM. The highest rate of regeneration, shoot number, leaf number, shoot length and dry weight of shoots were obtained from the nodal segments were inoculated into plant growth regulator free medium. The use of mannitol and sorbitol were not efficient for in vitro conservation catingueira, being suitable for this purpose, use of concentration of 219.10mM sucrose and 6μM PBZ until 240 days.

A cultura de tecidos vegetais tem-se mostrado eficaz na multiplicação e conservação de espécies lenhosas com potenciais explorados. Dessa forma, este trabalho objetivou estudar os efeitos dos tipos e concentrações de reguladores de crescimento vegetal sobre a multiplicação in vitro de C. pyramidalis e desenvolver um protocolo de conservação in vitro para a mesma. Para a multiplicação realizou-se três experimentos, onde foram avaliados os efeitos de diferentes concentrações dos reguladores de crescimento vegetal (BAP, KIN, TDZ e ANA) sobre diferentes explantes (hipocótilo, segmento cotiledonar, cotilédone, epicótilo e segmento nodal). Para a conservação in vitro, realizaram-se dois experimentos: no primeiro testou-se o efeito de diferentes concentrações de sacarose (87,64; 131,46; 175,28 e 219,10mM) combinados com concentrações (0,0 e 87,64mM) de sorbitol ou manitol, e no segundo, avaliou-se o uso de diferentes concentrações (0,0; 1,5; 3,0; 4,5 e 6,0µM) do retardante de crescimento paclobutrazol (PBZ). O meio de cultura usado para todos os experimentos foi o WPM. A maior taxa de regeneração, número de brotos, número de folhas, comprimento da parte aérea e matéria seca da parte aérea foram obtidas com o segmento nodal inoculado em meio isento de regulador de crescimento vegetal.
