Identificação e caracterização genômica de um isolado do Bean rugose mosaic virus

Resumo: The Bean rugose mosaic virus (BRMV) is a member of the genus Comovirus of the family Comoviridae, that infects leguminous plants. The complete genome of this virus had not yet been sequencenced. Comoviruses have two positive, single stranded genomic RNAs (RNA1 and RNA2), which are encapsidated into separate spherical virions. A virus isolate was mechanically inoculated in common bean cultivar Rio Tibagi, inducing symptoms of mosaic, blistering and leaf distortion. Indirect ELISA tests with antibodies for leguminous comoviruses were positives, and electron microscopic analysis revealed icosahedral particles approximately 28 nm in diameter in infected leaf samples. The virus transmission efficiency of Diabrotica speciosa was 40%. Reverse transcriptase-polymerase chain reaction (RT-PCR) analysis suggested the pathogen was a comovirus, as was confirmed after genomic partial sequencing. Eleven out of 29 soybean cultivars, and 17 out of 22 common bean cultivars showed symptoms after virus inoculation. The virus complete genome was sequenced consisting of two polyadenylated RNAs. RNA1 is 5909 nucleotides long encoding a single polypeptide of 1856 amino acids, with an estimated molecular weight of 210 kDa. The RNA1 polyprotein contains the polypeptides for viral replication and proteolytic processing. RNA2, that is 3644 nucleotides long, codes for a single 1097 amino acids polypeptide containing the movement protein and coat proteins. The complete nucleotide sequence of viral capsids (Small coat protein and Large coat protein) showed maximum identity of 54 and 61% with sequences from other Comovirus species. The isolate was identified as BRMV-Paraná, and based on sequencing data and on ICTV (International Committee on Taxonomy of Viruses) criteria, we propose BRMV as a distinct and definitive species in the Comovirus genus.

O genoma completo deste vírus ainda não havia sido sequenciado. Os comovírus possuem duas fitas simples de RNA de senso positivo (RNA1 e RNA2), que são encapsulados separadamente em vírions esféricos. Um isolado viral inoculado mecanicamente em feijoeiro cultivar Rio Tibagi, induziu sintomas de mosaico, associados à formação de bolhas e deformação foliar. Testes de ELISA indireto com anticorpos para comovírus de leguminosas foram positivos, e análises de microscopia eletrônica de transmissão revelaram partículas icosaédricas de aproximadamente 28 nm de diâmetro em amostras foliares infectadas. Nos testes de transmissão com Diabrotica speciosa, a eficiência de transmissão foi de 40%. Análises de transcrição reversa-reação em cadeia da polimerase (RT-PCR) sugeriram que o patógeno seria um comovírus, o que foi confirmado por análises de partes do genoma sequenciadas. Das 29 cultivares de soja, 11 apresentaram sintomas, e das 22 cultivares de feijão, 17 desenvolveram sintomas quando inoculadas com o vírus. O genoma completo do isolado viral foi sequenciado, sendo constituído de dois RNAs poliadenilados. O RNA1 é composto de 5909 nucleotídeos, codificando um único polipeptídeo com 1856 aminoácidos, e massa molecular estimada em 210 kDa. A poliproteína do RNA1 contém os polipeptídeos necessários para a replicação viral e para os processos proteolíticos. O RNA2, com 3644 nucleotídeos, codifica um único polipeptídeo de 1097 aminoácidos contendo uma proteína de movimento viral, e proteínas dos capsídeos. As sequências de nucleotídeos completas dos capsídeos virais (Small coat protein e Large coat protein) apresentaram identidade máxima de 54 e 61% com as sequências de outras espécies de Comovirus.
