Produção de anticorpos policlonais para a pesquisa de produção de toxina diftérica por cepas de Corynebacterium diphtheriae

Resumo: Despite the success of mass immunization in many countries, including in Brazil, diphtheria continues to be a potentially lethal resurgent infectious disease, which requires the use of diphtheria antitoxin (produced in horse) for treatment and diagnosis of toxin production. In this investigation the antiserum against diphtheria toxin was produced in rabbits and, after fractionation, immunoglobulin preparation was tested in parallel with horse antitoxin (SAD). Rabbits (n=2) were immunized with dimerized (non-toxic) diphtheria toxin. The experimental immunization was performed with consent of the University Experimental Animal Board (CEUA/033/2015). Clinical and reference strains were tested for the confirmation of species and toxin production by multiplex PCR. Reactivity against toxin produced by the strains was performed by phenotypic methods (Radial immunodiffusion and modified Elek tests). The capability of toxin neutralization was evaluated by VERO cell toxicity test. The detection of toxin in the surface of a toxin producer (ATCC 27012), and a non-toxin producer (ATCC 27010) strains was evaluated after biotinilation of Igs fraction, and revealed by fluorescence with streptavidin coupled to fluorescein. Horse antitoxin (SAD) presented many precipitation lines that can be mislead to toxin precipitin under modified Elek test, and many non-toxigenic strains gave positive results under radial immunodiffusion reaction analysis. An 80% discordance of toxin production result was observed by RID test amongst tox– strains, and 40% by Elek test, using SAD. The results with rabbit antibody fraction presented variable results for only one tox– strain (HC03). In general, a concordance of 90% was obtained by Elek test using rabbit antibody. The rabbit antibody fraction was capable to neutralize the toxin in a dilution of 1:1200 correlated to 0,1 IU of SAD. Preimmune sera of both rabbits were incapable to neutralize or react by Elek test. Biotinilated antibodies were capable to bind the tox+ ATCC 27012 strain, by fluorescence method. The rabbit antiserum rendered a more accurate reagent to identify tox+ C. diphtheriae strains, with a good neutralization titer. Biotinilated antibodies can be used for diagnosis of toxigenic C. diphtheriae strains

Nesta investigação, anticorpos antitoxina diftérica foram produzidos em coelhos e, após o fracionamento, a preparação de imunoglobulinas foram testados em paralelo à antitoxina produzida em cavalos (SAD comercial). Coelhos (n=2) foram imunizados com toxina diftérica dimerizada (desprovida de atividade tóxica). A imunização experimental nos coelhos foi realizada com o consentimento do comitê de experimentação animal da UERJ (CEUA/033/2015). As cepas clínicas e de referência foram testadas para a confirmação das espécies e produção de toxina pelo PCR multiplex. A reatividade contra a toxina produzida pelas cepas foi realizada pelos métodos fenotípicos usuais (imunodifusão radial simples e teste de Elek modificado). A capacidade de neutralização de toxina foi avaliada através do teste de citotoxicidade para células VERO. A detecção de toxina na superfície de amostra produtora de toxina (ATCC 27012) e não produtora de toxina (ATCC 27010) foi avaliada após a biotinilação da fração de Igs e por revelação com estreptavidina conjugada à fluoresceína. A antitoxina de cavalo (SAD) apresentou diversas linhas de precipitação que poderiam gerar confusão quando da leitura do ensaio, através do teste de Elek modificado e diversas cepas não toxinogênicas apresentaram resultados positivos através do ensaio de imunodifusão radial (IDR). Uma discordância de 80% foi observada nos resultados de IDR entre as amostras tox–, e de 40% através dos testes de Elek quando o SAD comercial foi utilizado. Os resultados utilizando a fração de imunoglobulinas de coelho apresentaram resultados variáveis apenas para uma cepa tox– (HC03). De modo geral, uma concordância de 90% foi obtida nos ensaios de Elek modificado utilizando anticorpos de coelho. A fração de anticorpos de coelho foi capaz de neutralizar a toxina na diluição 1:1200, quando comparado com 0.1 UI de SAD. O soro pré-imune de ambos os coelhos foram incapazes de neutralizar a toxina ou reagir através do teste de Elek. Os anticorpos biotinilados foram capazes de ligar à cepa tox+ ATCC 27012 através do método de fluorescência. O antissoro de coelho comportou-se como um reagente com maior precisão para a identificação das cepas tox+ de C. diphtheriae, com um bom título neutralizante.
