Análise comparativa das propriedades fenotípicas e genotípicas de amostras de origem humana e animal de Streptococcus pseudoporcinus

Resumo: The genus Streptococcus is constantly suffering reclassifications. The aim of this study was to evaluate the characteristics presented by 24 isolates of Streptococcus pseudoporcinus, including a derivative of the type strain, being 19 obtained from human clinical especimens and five from animal origin (milk samples). Six Streptococcus porcinus reference strains were included for comparative purposes. These isolates were characterized by physiological tests, protein profiles by SDS-PAGE, antimicrobial susceptibilities and virulence-related characteristics. Additionally, MALDI-TOF MS technique was used in order to characterize these isolates, allied with phylogenetic analysis and whole genome sequencing. Proteomic assays were carried out by mass spectrometry (ESI-Q-TOF). Physiological tests results indicated that hippurate hydrolysis was the only one able to differentiate the species. The results obtained by MALDI-TOF MS showed distinct scores values, depending on the protocol employed to the samples preparation. Protein extracts obtained by the treatment with formic acid and acetonitrile, have shown effective results in discriminate between S. pseudoporcinus and S. porcinus species. Additionally, high correlation degrees were identified among samples of S. pseudoporcinus obtained from milk. The data obtained by MALDI-TOF MS were also assessed to identify biomarkers, demonstrating the presence of intra and interspecies spectra. The analysis of the protein profiles obtained by SDS-PAGE revealed a great similarity between the species and it was not sufficiently discriminatory. The analysis by the rrs gene sequences that S. pseudoporcinus isolates belonging to human sources were less related to the isolates obtained from milk samples. Antimicrobial susceptibility testing demonstrated 66.0% of the S. pseudoporcinus of human origin were resistant to tetracycline. The isolates were able to form biofilms, and supplementation of the culture medium with sucrose or glucose considerably increases the amount of biomass adhered. Biofilms of type strains of both species showed inhibitory concentration values up to 40,000 times greater than planktonic cells. Several virulence-related proteins have been identified by proteomics and genomics analyses. Both species were able to adhere to human cells.

O objetivo deste estudo foi avaliar as características apresentadas por 24 amostras de Streptococcus pseudoporcinus, sendo 19 de origem humana, incluindo uma derivada da amostra tipo, e cinco de origem animal (isoladas de leite), em comparação com amostras de referência de Streptococcus porcinus. Estas amostras foram caracterizadas por testes fisiológicos, perfil total de proteínas por SDS-PAGE, susceptibilidade a antimicrobianos e atributos relacionados a virulência. Além disso, a técnica de MALDI-TOF foi utilizada na caracterização das amostras, aliado a análise filogenética e genômica comparativa in silico. Também foi analisado o perfil proteômico utilizando espectrometria de massa (ESI-Q-TOF). Dos testes fisiológicos utilizados, a hidrólise do hipurato foi o único capaz de diferenciar as espécies. Na metodologia de MALDI-TOF, os resultados obtidos revelaram valores de escores distintos dependendo do tipo do protocolo empregado. Extratos proteicos obtidos pelo tratamento com ácido fórmico e acetonitrila, demonstraram resultados eficazes. A partir dos espectros obtidos, foi determinada uma similaridade entre as amostras do estudo. Houve uma perfeita discriminação entre as amostras S. pseudoporcinus das de S. porcinus. Adicionalmente, as amostras de S. pseudoporcinus obtidas de leite mostraram uma maior correlação entre si. Os dados obtidos com a metodologia de MALDI-TOF foram também avaliados quanto à identificação de biomarcadores, demonstrando a presença de picos intra e inter-específicos. A análise dos perfis proteicos obtidos por SDS-PAGE revelou uma grande similaridade entre as espécies, não tendo sido suficientemente discriminatória. A análise pelo pareamento das sequências do gene rrs, disponibilizadas em bancos de dados públicos, revelou que as amostras de S. pseudoporcinus de fonte humana estão menos relacionadas com as amostras de fonte animal. Os testes de susceptibilidade aos antimicrobianos demonstraram 66,0% de amostras de S. pseudoporcinus de origem humana resistentes à tetraciclina. Todas as amostras se mostraram capazes de formar biofilmes, sendo que a suplementação do meio de cultura com sacarose ou glicose aumenta consideravelmente a quantidade de biomassa aderida. As amostras tipo de ambas as espécies, quando cultivadas sob a forma de biofilmes apresentaram valores de concentração inibitória até 40.000 vezes maiores que células planctônicas. Diversas proteínas relacionadas a virulência foram identificadas pelas análises proteômica e genômica. Ambas as espécies foram capazes de aderir a células humanas.
