Resumo: | According to WHO, smoking kills more than seven million people each year and lead to more than $ 1.4 trillion in expenses by governments. Despite widespread disclosure of cigarette maltreatment during pregnancy, it is estimated that 20-23% of women in Brazil smoke during gestation. Although the neurotoxic effects of cigarette use on brain development are known, the mechanisms involved in this process remain largely unknown. Considering that the visual system is widely recognized as a model of neural circuitry used in the study of the development and plasticity of nerve connections, the investigation of the effects of cigarette use on the development of the visual system offers great potential to elucidate mechanisms related to deleterious effects during pregnancy. Thus, the central hypothesis of this study was that the exogenous introduction of nicotine and other components of cigarette smoke would interfere with the development and maturation of nerve connections in the visual system. Specifically, we evaluated the impact of pre-and peri-natal exposure of mice to smoking and oral nicotine on the development of retinal connections to the dorsal lateral geniculate nucleus (dLGN), superior colliculus (SC), and olivary pretectal nucleus (OPN). Intraocular injections of the Alexa Fluor fluorophores conjugated cholera toxin B (CTB) subunit were done to visualize retinal ganglion cell projections in the dLGN, SC and OPN. Overall, our findings revealed that exposure to cigarette smoke or to nicotine via oral administration during development have a profound effect on the refinement of retinal projections in the studied regions. Moreover, such exposure led to an increase in the incidence of visual abnormalities, loss of body weight and early eye opening. Thus, we believe that our results confirm our hypothesis that the exogenous introduction of nicotine and other components of cigarette smoke interfere with the development and maturation of retinal connections in the visual system of mice. Moreover, our findings contribute to the vast literature on the adverse consequences of smoking exposure during brain development, which currently afflicts thousands of children in Brazil. |

Além disso, é estimado que 20-23% das mulheres no Brasil fumem durante a gestação. Desta forma, é revelante investigar os efeitos neurotóxicos do cigarro no desenvolvimento cerebral, pois muitos mecanismos deste processo são desconhecidos. Sabe-se que o sistema visual é amplamente reconhecido e utilizado como modelo de circuito neuronal no estudo do desenvolvimento e na plasticidade das conexões nervosas. Sendo assim, este trabalho utilizou o sistema visual como modelo de estudo para averiguar os possíveis efeitos da exposição pré e pós-natal à fumaça do cigarro no desenvolvimento das conexões retinotalâmicas e retinocoliculares de camundongos. Camundongos C57Bl/6 foram expostos à nicotina por exposição à máquina de fumaça do cigarro (SMK, 0,73mg de nicotina/cigarro) ou por oferta em solução oral (NIC ORAL, 50µg/mL adoçada sacarina à 2%). Seus respectivos grupos controles foram expostos ao ar (CTRL AR) ou a oferta de água (CTRL ÁGUA). Os animais dos grupos SMK TOTAL e NIC ORAL foram expostos durante todo período de desenvolvimento embrionário e nas duas primeiras semanas de vida pós-natal, assim como os respectivos controles. Já os animais do grupo SMK PARCIAL foram expostos somente nas duas primeiras semanas de vida pós-natal (PN2-16). Entre os dias pós-natal 11-14 (±PN12), os camundongos receberam injeções intraoculares de uma solução contendo a subunidade B da toxina da cólera (CTB) conjugada a fluoróforos com comprimento de onda 488nm ou 594nm, para a visualização das projeções da retina no tálamo visual, colículo superior (CS) e núcleo olivar pretectal (NOP). A quantificação das projeções retinianas no núcleo geniculado lateral dorsal (NGLd), CS e NOP foi efetuada através de medidas das extensões espaciais das projeções de cada olho. O padrão de marcação das projeções retinotalâmicas no NGLd indicou que animais expostos à nicotina, seja por fumaça do cigarro ou via oral, exibiram uma maior sobreposição entre as projeções contra e ipsolaterais. Do mesmo modo, esses animais apresentaram maior comprimento nas projeções ipsolaterais no CS. Contudo, não foram observadas alterações no NOP de animais expostos à fumaça do cigarro ou nicotina via oral. Além disso, as proles de progenitoras expostas à fumaça de cigarro apresentaram 4 vezes mais anomalias visuais do que os animais controles. Também foram observados redução na massa corpórea dos animais expostos à fumaça do cigarro e a nicotina por via oral.
