Estudo dos potenciais citotóxico, antineoplásico e mutagênico do extrato etanólico de Hovenia dulcis Thunberg

Resumo: Hovenia dulcis Thunberg, is popularly known as Japanese grape, occurring naturally in China, Japan and Korea. The species has many biological activities, highlighting the antineoplastic activity on several cell types. This work aims to evaluate the cytotoxic, antineoplastic and mutagenic potential of the ethanolic extract of Hovenia dulcis leaves. H. dulcis leaves were collected at the Clube dos Médicos in Petropólis (22 ° 30'01.1 "S 43 ° 13'32.8" W), in October 2015. The plant extract used in this work was prepared from leaves immersed in 95% ethanol. Cells of lineage A549 were treated with different concentrations of this extract. After the 24-hour treatment period, cytotoxic activity was analyzed by the methods of WST-1, trypan blue exclusion and clonogenic recovery. The dose that inhibits 50% of cell growth was verified when the cells were exposed to the extract in concentrations of 250 and 275 μg / mL. These concentrations were tested in the normal Vero strain. In this strain, a 10% decrease in cell viability was observed through the trypan blue exclusion tests and 9% in the WST- 1 assay. In the clonogenic recovery assay, there was no statistically significant decrease in the mitogenic capacity of the cells when compared to the control. Experiments were carried out to evaluate the production rate of reactive oxygen species, using flow cytometry using the DCFH-DA reagent, where the extract was shown to be an antioxidant for both tested strains. The yeast Saccharomyces cerevisiae FF18733 (wild) and CD138 (mutant) were also used as a biological model in this study. Both were evaluated for survival fraction, growth, mutagenesis (canavanin resistance test) and petite colonies. And the results showed that the concentration of 150 μg / mL and 100 μg / mL decreased cell viability by 50% in strains FF18733 and CD138 respectively. In the growth test the strains were exposed for 48 hours to the extract of H. dulcis. There was no statistically significant decrease in the FF1873 strain, when compared to the control, whereas the CD138 strain, there was a significant difference in the growth rate. However, in the canavanin resistance test, H. dulcis extract was mutagenic for both strains. In the experiments to evaluate the loss of mitochondrial function, it was observed that in both strains tested they presented an increase in the rate of petite colonies. Comparing the results observed in the upper and lower eukaryotes, we observed that in both cases there was an alteration in the oxidative pathways that affect the mitochondrial metabolism, suggesting that this organelle is the most affected by the products of the extract of H. dulcis, being necessary a study analyzing the pathway cell death, and about the chemical composition of the extract.

A espécie apresenta muitas atividades biológicas, destacando-se a atividade antineoplásica sobre diversos tipos celulares. Este trabalho visa avaliar a potencialidades citotóxica, antineoplásica e mutagênica do extrato etanólico de folhas de Hovenia dulcis. Folhas de H. dulcis, foram coletadas nas dependências do Clube dos médicos em Petropólis (22°30'01.1"S 43°13'32.8"W), no mês de outubro de 2015. O extrato vegetal utilizado neste trabalho foi preparado a partir de folhas imersas em etanol 95%. Células da linhagem A549 foram tratadas com diferentes concentrações desse extrato. Após o período de tratamento de 24 horas, a atividade citotóxica foi analisada pelos métodos de WST-1, exclusão por azul de tripan e recuperação clonogênica. Verificou-se a dose que inibe 50% do crescimento celular, quando as células foram expostas ao extrato nas concentrações de 250 e 275 μg/mL. Essas concentrações foram testadas na linhagem normal Vero. Nesta linhagem, observou-se diminuição de 10% na viabilidade celular através dos testes de exclusão por azul de tripan e 9% no ensaio de WST-1. No ensaio de recuperação clonogênica, não se observou diminuição estatisticamente significativa, na capacidade mitogênica das células quando comparadas ao controle. Realizou-se experimentos para avaliar a taxa de produção de espécies reativas de oxigênio, através de citometria de fluxo utilizando o reagente DCFH-DA, onde o extrato se mostrou antioxidante para ambas linhagens testadas. Utilizou-se a levedura Saccharomyces cerevisiae FF18733 (selvagem) e CD138 (mutante) como modelo biológico neste estudo, as quais foram avaliadas quanto à fração de sobrevivência, crescimento, mutagênese (ensaio de resistência a canavanina) e colônias petites. Os resultados apontaram que a concentração de 150 μg/mL e 100 μg/mL diminuíram a viabilidade celular em 50% nas cepas FF18733 e CD138. No ensaio de crescimento as cepas foram expostas por 48 horas ao extrato de H. dulcis. Não foi observado diminuição estatisticamente significativa na cepa FF1873, quando comparada ao controle. E a cepa CD138, apresentou diferença significativa na taxa de crescimento. Entretanto no ensaio de resistência a canavanina, o extrato de H. dulcis se mostrou mutagênico para ambas as cepas. Nos experimentos para avaliar a perda da função mitocondrial, observouse que as duas cepas apresentaram aumento da taxa de colônias petites. Ao comparar os resultados observados em eucarioto superior e inferior, percebeu-se que ocorreu uma alteração nas vias oxidativas, que afetam o metabolismo mitocondrialnos nos dois casos.
