Interação de azul de metileno com modelos de membranasTERAPIA FOTODINÂMICAAZUL DE METILENODINÂMICA MOLECULARPHOTODYNAMIC THERAPYMETHYLENE BLUEMOLECULAR DYNAMICSPROGRAMA DE PÓS-GRADUAÇÃO EM NANOCIÊNCIAS E MATERIAIS AVANÇADOS - UFABCOrientador: Prof. Dr. Ronei MiottoTese (doutorado) - Universidade Federal do ABC, Programa de Pós-Graduação em Nanociências e Materiais Avançados, 2015.Neste trabalho, estudamos o processo de interação de fotossensibilizadores com modelos de membranas lipídicas com o intuito de contribuir para a compreensão dos processos dinâmicos e das mudanças físicas e estruturais em nível microscópico. Em particular, nossa análise está voltada para o desenvolvimento de fotossensibi- lizadores mais eficientes para aplicação em terapia fotodinâmica (TFD). Nossos estudos baseiam-se principalmente em simulações atomísticas de dinâmica molecu- lar (MD). A topologia do fotossensibilizador alvo, azul de metileno (AM), é obtido através de um modelamento quântico, baseado na teoria do funcional da densidade (DFT). Todas as simulações MD iniciam-se com o AM disperso em fase aquosa em diferentes configurações. Independente da configuração inicial, o AM interage com as membranas modelo estudadas em aproximadamente 2 ns. O processo de intera- ção é mediado principalmente por interações eletrostáticas entre membrana e AM. Nossos cálculos indicam que todas as membranas modelos estudadas apresentam modificações na presença de AM, mesmo na ausência de irradiação, sugerindo que algumas das mudanças morfológicas da membrana observadas experimentalmente acontecem antes do processo de iluminação. De acordo com nossas simulações, as membranas estudadas apresentam um aumento de área por lipídeo e uma diminui- ção da espessura, indicando um aumento da permeabilidade da membrana o que implica em uma maior facilidade para trocas entre os meios interno e externo. A análise da estrutura atômica derivada de nossas simulações sugerem que o átomo de enxofre da molécula de azul de metileno interage principalmente com os fosfatos presentes nas cabeças polares dos fosfolipídeos que compõem as membranas mo- delo. Nossas simulações indicam que o grau de superposição do AM com o oxigênio dissolvido no meio é similar para todas as membranas modelo consideradas. Isso nos leva a inferir que a eficiência do AM como agente em TFD nos diferentes mode- los de membrana considerados depende muito mais da afinidade entre membrana e fotossensibilizador do que das características estruturais ou modificações provo- cadas pela internalização do AM. Finalmente, cabe destacar que nossos cálculos indicam que a presença de íons no solvente provocam mudanças nas características de penetração do AM dificultando a passagem da molécula nos casos da DPPC, POPC e POPG.In this work, we study the photosensitizers interaction process with model lipid membranes in order to contribute to the understanding of the dynamic processes with physical and structural changes at the microscopic level. In particular, our analysis is focused on the development of more efficient photosensitizers for use in photodynamic therapy (PDT). Our studies are mainly based on atomistic simu- lations of molecular dynamics (MD). The topology of the target photosensitizer, methylene blue (MB), is obtained by a quantum modeling based on Density Func- tional Theory (DFT). All MD simulations begin with MB in the dispersed aqueous phase in different configurations. Regardless of the initial configuration, the MB interacts with model membranes studied in approximately 2 ns. The process of interaction is primarily mediated by electrostatic interactions between membrane and MB. Our calculations indicate that all membranes studied models show chan- ges in the presence of MB, even in the absence of irradiation, suggesting that some of the experimentally observed morphological changes occur before the membrane illumination process. According to our simulations, we studied the membranes have an area of increased lipid and a thinning, indicating an increased permea- bility of the membrane which implies a greater ability to exchange between the internal and external means. The analysis of atomic structure derived from our simulations suggest that the sulfur atom the methylene blue molecule interacts mainly with phosphates in the polar heads of the phospholipids that makes up the model membranes. Our simulations indicate that the MB degree of overlap with the dissolved oxygen in the medium is similar for all the considered model membranes. This leads us to infer that the MB efficiency as an agent in PDT in different models of membrane considered depends much more on affinity between membrane and photosensitizer that the structural characteristics or modifications caused by internalization of MB.
