Juntas, acumulam 100 mil casos anuais, atingindo principalmente populações vulneráveis de países em desenvolvimento. Considerando a sua letalidade e os importantes problemas de toxicidade associados aos atuais tratamentos, é urgente a busca por novas substâncias ativas contra estas doenças. Com respeito ao câncer, sendo esta a segunda maior causa de morte no mundo, a prospecção de novos quimioterápicos é imprescindível. Especificamente, o câncer de cabeça e pescoço tem na quimioterapia uma de suas principais estratégias de tratamento, visando a cura e a preservação da funcionalidade dos órgãos acometidos. No contexto dessas problemáticas, e partindo da premissa de que os metabólitos especiais oriundos de plantas são fonte inesgotável de substâncias bioativas, o presente trabalho traz um estudo comparativo de atividades anti-T. cruzi , anti-L. (L.) infantum e antitumoral entre o alcaloide aporfínico (S)-dicentrina, obtido de O. puberula, e de três derivados: (S)-N-metildicentrina (1), (6aS, 6S)-dicentrina N-óxido (2) e (6aS, 6R)-dicentrina N-óxido (3), sendo este último um composto inédito. Todos os compostos obtidos foram caracterizados por técnicas espectroscópicas e espectrométricas. O derivado N-oxidado (2) foi o mais ativo frente à forma amastigota de T. cruzi, (CE50 = 9,9 ?M e IS > 20,2; benzonidazol: CE50 = 6,5 ?M e IS > 30,7), indicando melhora na atividade e na seletividade do alcaloide de partida. Frente à forma amastigota de L. (L.) infantum, o N-óxido (3) foi o derivado mais ativo (CE50 = 34,5 ?M; miltefosina: CE50 = 10,2 ?M; (S)-dicentrina: CE50 = 10,3 ?M), apresentando melhor IS do que seu precursor (IS > 5,8; (S)-dicentrina: IS = 5). Frente às linhagens tumorais SCC-143 e SCC-154, associadas com o câncer de cabeça e pescoço, o composto de partida foi mais ativo (CI50 = 7,1 e 20,0 ?M, respectivamente; cisplatina: CI50 = 3,6 e 0,8 ?M). Em geral, a N-oxidação mostrou ser uma funcionalização mais promissora que a N-metilação, gerando compostos mais ativos e menos tóxicos nos modelos biológicos estudados. Além disso, a estereoquímica destes derivados se apresentou como uma característica essencial para expressão de atividade.Chagas disease and visceral leishmaniasis are serious neglected tropical diseases caused, respectively, by the protozoa Trypanosoma cruzi and Leishmania (L.) infantum. Together, they accumulate 100,000 annual cases, mainly affecting vulnerable populations in developing countries. Considering its lethality and the important toxicity problems associated with current treatments, it is urgent to search for new active substances against these diseases. With regard to cancer, which is the second leading cause of death in the world, prospecting for new chemotherapy is essential. Specifically, chemotherapy is one of the main treatment strategies for head and neck cancer, aiming at curing and preserving the functionality of the affected organs. In this context, and based on the premise that special metabolites derived from plants are an inexhaustible source of bioactive substances, the present work presents a comparative study of anti-T.cruzi, anti-L. (L.) infantum and antitumoral among the aporphine alkaloid (S)-dicentrine, obtained from O. puberula, and three derivatives: (S)-N-methyldicentrine (1), (6aS, 6S)-dicentrine N-oxide (2) and (6aS, 6R)-dicentrine N-oxide (3), the latter being a novel compound. The four compounds obtained were characterized by spectroscopic and spectrometric techniques. The N-oxide derivative (2) was the most active against the amastigote form of T. cruzi, (CE50 = 9.9 ?M and SI > 20.2; benznidazole: CE50 = 6.5 ?M and SI > 30.7), indicating improvement on the activity and selectivity of the starting alkaloid. Against the amastigote form of L. (L.) infantum, N-oxide (3) was the most active derivative (EC50 = 34.5 ?M; miltefosine: EC50 = 10.2 ?M; (S)-dicentrine: EC50 = 10.3 ?M), showing better SI than its precursor (SI > 5.8; (S)-dicentrine: SI = 5). Against the SCC-143 and SCC-154 tumor cell lines, associated with head and neck cancer, the starting alkaloid was more active (IC50 = 7.1 and 20.0 ?M, respectively; cisplatin: IC50 = 3.6 and 0.8 ?M). In general, N-oxidation showed to be a more promising functionalization than N-methylation, generating more active and less toxic compounds in the biological models studied.
