Ana Lígia ScottCoorientador: Prof. Dr. Eric Allison PhilotTese (doutorado) - Universidade Federal do ABC, Programa de Pós-Graduação em Biossistemas, Santo André, 2022.O hDAT é uma proteína que relaciona-se com regulação da concentração de dopamina (DA) na fenda sináptica, permitindo o transporte do meio extracelular para o meio intracelular, sendo finamente regulada por modificações pós-traducionais que podem modular sua função, entre umas delas a fosforilação. O hDAT está associado com doença de Parkinson, dependência de substâncias psicoativas, entre outras doenças. O objetivo desta Tese é investigar os mecanismos moleculares envolvidos na regulação do transportador de dopamina nos terminais nervosos. Inicialmente foi necessário construir um modelo do hDAT e foi utilizado para gerar estruturas com diferentes padrões de fosforilação (P-22, NP-12, NP-333 e NP-all) que foram usadas para calcular a amostragem conformacional utilizando métodos híbridos (VMOD e MDeNM). Posteriormente, os dados foram analisados usando as cadeias de Markov para obter os perfis de energia livre, a identificação dos meta estados neles e para o cálculo das probabilidades de transição entre os meta-estados. Entre os principais resultados, identificou-se que alterações no padrão de fosforilação no N-terminal e no C terminal do hDAT podem alterar a amostragem dos meta-estado necessários para o transporte de DA. Além disso, estas alterações impactam os movimentos e os modos normais de baixa frequência, alterando com isto a dinâmica estrutural do hDAT. Também foi investigado como os diferentes padrões de fosforilação podem influenciar na passagem da dopamina pelo canal do hDAT tanto no influxo como no efluxo. Nesta parte do estudo foi empregado o método Steered Molecular Dynamics com o qual foi possível sugerir quais padrões de fosforilação podem aumentar ou diminuir a passagem do DA tanto no influxo como no efluxo. Como um dos resultados, identificamos que o sistema NP-12 possui uma menor resistência no influxo de dopamina, o que sugere uma maior taxa de transporte, corroborando estudos experimentais. Como conclusão pode-se destacar que mudanças na fosforilação de um resíduo induz a alterações na dinâmica estrutural do hDAT, podendo dificultar os diferentes meta-estados associados com o transporte da DA. Essa alteração na função de transporte pode afetar a homeostase de DA em neurônios dopaminérgicos, impactando a sobrevida destas células.hDAT is a protein that is related to the regulation of dopamine (DA) concentration in the synaptic cleft, allowing the transport from the extracellular environment to the intracellular environment, being finely regulated by post-translational modifications that can modulate its function, among them phosphorylation. hDAT has been associated with Parkinson's disease, psychoactive substance dependence drugs, among other diseases. The aim of this Thesis is to investigate the molecular mechanisms involved in the regulation of the dopamine transporter in nerve endings. Initially, a model of hDAT had to be built and was used to generate structures with different phosphorylation patterns (P-22, NP-12, NP-333 and NP-all) that were used to calculate conformational sampling using hybrid methods (VMOD and MDeNM). Subsequently, the data were analyzed using Markov chains to obtain the free energy profiles, the identification of the meta-states in them, and to calculate the transition probabilities between the meta-states. Among the main results, it was identified that changes in the phosphorylation pattern at the N-terminal and C-terminal of hDAT can alter the sampling of the meta states required for DA transport. Furthermore, these changes impact the normal low frequency motions and modes, thereby altering the structural dynamics of hDAT. It was also investigated how different phosphorylation patterns can influence the passage of dopamine through the hDAT channel in both influx and efflux. In this part of the study the Steered Molecular Dynamics method was employed with which it was possible to suggest which phosphorylation patterns can increase or decrease the passage of DA in both influx and efflux. As one of the results, we identified that the NP 12 system has a lower resistance in dopamine influx, which suggests a higher rate of transport, corroborating experimental studies. As a conclusion one can highlight that changes in the phosphorylation of a residue induces changes in the structural dynamics of hDAT, and may hinder the different meta-states associated with DA transport.
