Indução de bactérias olubilizadoras de fosfatos em cultivo de feijão-caupi (Vigna unguiculata (L.) Walp.)

Resumo: | Ecological relationship between microorganisms and plants is well known, however it is still under-explored. Thus, the aim of this research was to evaluate bacterial consortia as a growth inducers on cultivation of cowpea (Vigna unguiculata (L.) Walp.) by phosphate-solubilizing bacteria (PSB) of rizospheric soil of a cactus from Caatinga biome. Soil samples were analyzed and quantified to Cultivable Heterotrophic Bacteria (CHB) and PSB by Standard Plate Count (SPC) on Plate Count Agar (PCA) and Yeast Dextrose Agar (YDA), respectively. Twelve strains were isolated and submitted to Gram stain and Phosphate-Ferric (FePO4) solubilization capacity test. Antagonistic test by cross-streak technique was performed to assembly of bacterial consortia. Bacterial consortia was introduced on cultivation of cowpea, which was performed on vegetable house of Departamento de Fitotecnia, Universidade Federal do Ceará. After 45 cultivation days, the plants were evaluated to aerial portion length (APL), aerial portion dry mass (APDM), root dry mass (RDM) and total dry mass (TDM). The substrate was evaluated to labile P content. Variables were submitted to statistical tests. CHB and PSB counts showed values of 4.62×107 and 3.475×107 CFU/g, respectively. About 82% of strains were Gramnegative and 18% were Gram-positive. Of the 12 strains capable to solubilize CaHPO4 seven were capable to solubilize FePO4. From the antagonism test were elected four bacterial consortia which were induced on cowpea cultivation. According to statistical tests, C4 consortia was more efficient by BDM and TDM accumulation. From the labile P substrate accumulation analysis, C3 and C4 consortia were showed best efficiency to this process. Therefore, it was possible to verify that the treatments with higher TDM averages were also those with higher labile P content in the substrate. |

Dessa forma, o objetivo desta pesquisa foi testar consórcios bacterianos como indutores de crescimento em cultivo de feijão-caupi (Vigna unguiculata (L.) Walp.) formados a partir de isolamentos de bactérias solubilizadoras fosfato (BSF) do solo rizosférico de uma cactácea do bioma Caatinga. A amostra de solo foi analisada e foram quantificadas bactérias heterotróficas cultiváveis (BHC) e BSF através do método de contagem padrão em placas (CPP), utilizando ágar padrão de contagem (Agar PCA) e ágar extrato de levedura e glicose (Agar GL), respectivamente. Foram isoladas 12 cepas e submetidas ao teste de coloração de Gram para caracterização morfotintorial e testes de capacidade de solubilização de fosfato férrico (FePO4). O teste de antagonismo pela técnica de estrias cruzadas foi usado como critério na montagem dos consórcios bacterianos. Os consórcios foram introduzidos em cultivo de feijão-caupi realizado em casa de vegetação do departamento de Fitotecnia da Universidade Federal do Ceará. Passados 45 dias de cultivo, as plantas foram avaliadas quanto ao comprimento da parte aérea (CPA); massa seca da parte aérea (MSPA); massa seca da raiz (MSR); e massa seca total (MST). O substrato foi avaliado quanto ao teor de P lábil. As variáveis foram submetidas a teste estatístico. As contagens de BHC e BSF apresentaram valores de 4,62×107 e 3,475×107 UFC/g, respectivamente. Cerca de 82% dos isolados apresentaram morfologia de bastonetes Gram-negativos e 18% de bastonetes Gram-positivos. Dos 12 isolados capazes de solubilizar CaHPO4 sete foram capazes de solubilizar FePO4. A partir do teste de antagonismo foi eleito quatro consórcios bacterianos que foram induzidos no cultivo de feijão-caupi. Pode-se inferir, pela análise estatística, que o consórcio C4 foi mais eficiente que os demais no acúmulo de MSR e MST. Da análise de acúmulo de P lábil no substrato, C3 e C4 foram os que apresentaram melhor eficiência para esse processo.
