Resumo: An important mechanism for survival cell, carried out by the plant species response to adverse or stressful situations, is the accumulation of proteins, especially the soluble proteins as well as amino acids and other nitrogen compounds. In this study, was determined distribution of nitrogen compounds during heat stress in different parts of the plant. It can be observed that with increasing temperature of 25 °C to 45 °C, there was greater accumulation of soluble fraction in leaves (40%), while the increase in the insoluble fraction was approximately 20% while the accumulation of proline was about five times in the leaves of cashew while this increase in the roots was only once. The concentration of total free amino acids was high both in leaves (5%) and roots (9%). The content of ammonium has increased both in roots (30%) and leaves (60%) with increasing temperature of 25 °C to 45 °C. Increasing the protein content in the cashew leaves, induced by heat is related to the mobilization of nitrogen reserves from roots and stems and/or protein synthesis in leaf tissue. Based on these observations, in addition to protein deposition during heat stress, we identified specific patterns of protein expression of cashew against specific and combined to abiotic stresses. The protein pattern in cashew leaves was compared by twodimensional electrophoresis (2D) after to the treatments: control conditions, irrigated with Hoagland & Arnon solution diluted to ¼, or subjected to drought by suspension of irrigation for 20 days, and then both the control seedlings as the pre-exposed to drought were subjected to 25 °C or 45 °C. The protein expression changes were in 80 proteins leaves of cashew. In the condition of heat, 7 protein increased the expression and 3 proteins were apparently synthesized de novo. In dry condition, 10 proteins increased their expression and 2 proteins were identified as plastocyanin and ferredoxin. The analysis also showed that 17 proteins were altered by the effect of the stress combination and as well as heat stress, 14 increased expression and 2 were found only in the stress combination. Thus, our results suggest that the stresses combination should be considered as a new state of abiotic stress in plants, and not only as a combination of abiotic factors.

Silveira, Joaquim Albenísio Gomes daLima, Cristina Silva de2015-01-15T18:20:39Z2015-01-15T18:20:39Z2009info:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/masterThesisapplication/pdfLIMA, C.
