Estudo etnofarmacológico e screening de atividades farmacológicas do óleo essencial extraído das folhas de Croton cordiifolius baill

Resumo: The use of plants has derived its principles from traditional medicine. Croton genus is known for the presence of species rich in biologically active secondary metabolites. Croton cordiifolius Baill. is a shrub known as “quebra faca” found in the state of Pernambuco and used by local people for various purposes . However, there are no studies to prove its effects. This study started by an ethnobotanical survey in a rural area of semiarid region of Pernambuco, in which “quebra faca” was cited mainly as anti-inflammatory and wound healing. So, it was decided to assess the acute toxicity, topical anti-inflammatory, wound healing, antibacterial and antinociceptive activities of the essential oil extracted from the fresh leaves of this species. Specimens of C. cordiifolius were collected, and a voucher specimen deposited in the herbarium of the Agronomic Institute of Pernambuco. The essential oil was extracted by hydrodistillation and its composition determined by gas chromatography to a mass spectrometry gas. For in vivo testing mice (2- 4 months) and for in vitro antimicrobial activity, clinical isolated from wounds were used. Chemical analysis of the essential oil showed, as major constituents, 1,8-cineole and -phellandrene. The essential oil from Croton cordiifolius (EOCc) showed low toxicity (LD50 > 1000 mg/kg) , topical anti-inflammatory activity in the ear edema induced by croton oil and arachidonic acid as well as in excisionial wounds, in which also stimulated angiogenesis and collagen deposition. Regarding antinociceptive activity, EOCc reduced the number of writhing induced by acetic acid and the licking time in both phases of the formalin test, showing its antinociceptive effect, with no involvement of opioid system and vanilloid receptors (TRPV-1), and participation of the glutamatergic system. These data show the pharmacologic potential of this specie in wound healing process, as well as in pain and inflammation reduction.

O gênero Croton é conhecido pela presença de espécies ricas em metabólitos secundários biologicamente ativos. Croton cordiifolius Baill. é um arbusto conhecido como quebra faca encontrado no interior de Pernambuco e utilizado pela população local para diversos fins. Contudo não há estudos que comprove seus efeitos. O presente estudo iniciou-se por um inquérito etnobotânico realizado na área Rural do semiárido Pernambucano, ocasião em que o quebra faca foi citado principalmente como anti-inflamatório e cicatrizante. Portanto foi decido avaliar a toxicidade aguda, atividade anti-inflamatória tópica, cicatrizante, antimicrobiana e antinociceptiva do óleo essencial extraído das folhas frescas desta espécie. Espécimes de C. cordiifolius foram coletados, e uma exsicata depositada no herbário do Instituto Agronômico de Pernambuco. O óleo essencial foi extraído por hidrodestilação e sua composição determinada por cromatografia a gás acoplada a espectrômetro de massa. Para os ensaios in vivo foram utilizados camundongos (2-4 meses) e para atividade antimicrobiana in vitro cepas de bactérias isoladas de feridas. Análise química do óleo essencial evidenciou, como constituintes majoritários, o 1,8-cineol e o -felanfreno. O óleo essencial de Croton cordiifolius (OECc) apresentou baixa toxicidade (DL50 >1000 mg/kg), atividade anti-inflamatória tópica no edema de orelha induzido por óleo de Cróton e ácido araquidônico, bem como em feridas excisioniais. Nas quais também estimulou a angiogênese e deposição de colágeno. Quanto a atividade antinociceptiva, o OECc promoveu redução do número de contorções abdominais por ácido acético e no tempo de lambida de pata em ambas as fases do teste da formalina, evidenciando seu efeito antinociceptivo, que se mostrou independente da via dos opioides e dos receptores TRPV-1, com participação do sistema glutamatérgico.
