Withanolides isolated from the leaves extract of Acnistus arborescens

Resumo: This paper describes the qualitative analysis of the constituents of ethanol extract from leaves of Acnistus arborescens grown in the NÃcleo de Estudos e Pesquisas em Agricultura Urbana (NEPAU), Departamento de Agronomia da Universidade Federal do Cearà (UFC). This study resulted in the development of an analytical method for the isolation and characterization of steroidal lactones, known as withanolides, from the analysis of the extract using chromatographic methods, such as column chromatography (CC), thin layer chromatography (TLC) and high-performance liquid chromatography (HPLC). The isolated compounds had their structures determined by the application of spectroscopic techniques such as nuclear magnetic resonance of hydrogen-1 and carbon-13 (one and two-dimensional) and absorption in the infrared. Thus, it was possible to isolate and identify two vitanolidos previously described in the literature, these being known as Phyalolactone and Withanolide D, and two others withanolides that, according to the literature review done, are considered unique in the literature.

Este estudo resultou no desenvolvimento de um mÃtodo analÃtico para isolamento e caracterizaÃÃo de lactonas esteroidais, conhecidas como vitanolÃdeos, a partir da anÃlise do extrato por meio de mÃtodos cromatogrÃficos, como: Cromatografia em Coluna (CC), Cromatografia em Camada Delgada (CCD) e Cromatografia LÃquida de Alta EficiÃncia (CLAE). As substÃncias isoladas tiveram suas estruturas determinadas atravÃs da aplicaÃÃo de tÃcnicas espectroscÃpicas como: RessonÃncia MagnÃtica Nuclear de hidrogÃnio-1 e carbono-13 (uni e bidimensional) e absorÃÃo na regiÃo do infravermelho. Desta forma, foi possÃvel o isolamento e identificaÃÃo de dois vitanolÃdeos jà descritos anteriormente na literatura, sendo estes conhecidos como Fisalolactona e VitanolÃdeo D, alÃm de dois outros vitanolÃdeos os quais, de acordo com o levantamento bibliogrÃfico feito, sÃo considerados inÃditos na literatura.
