Efeitos do treinamento físico sobre a função cardíaca de ratas ovariectomizadas após o infarto do miocárdio

Resumo: Approximately 40% of patients with myocardial infarction develop ventricular dysfunction with or without signs of heart failure, which adversely affect the quality of life, rates of hospitalization and mortality. The adverse ventricular remodeling after an acute ischemic event is the main factor involved in ventricular dysfunction and presents important differences between males and females. Studies have shown that the reduction of ovarian hormones can interfere negatively in important pathways related to the remodeling process. In addition, the regular practice of exercise training have shown important beneficial effects after myocardial infarction. Therefore, the aims of this study were to analyze the effects of exercise training in treadmill on the cardiac function, histologic and morphometric parameters and to check the protein expression of cardiac renin-angiotensin, oxidant and antioxidant systems in ovariectomized rats after myocardial infarction. Female Wistar rats (Rattus Norvegicus Albinus), with eight weeks of age were randomly divided into five groups at the time of ovariectomy: Control, OVX+SHAMSED, OVX+SHAMEF, OVX+IMSED and OVX+IMEF. Two weeks after the induction of myocardial infarction animals started the exercise training. The training protocol was carried out for a period of eight weeks, with duration of 60 min/day, five times a week with an interval of two days to rest. Fortyeight hours after the last day of exercise training protocol, animals were anesthetized for the achievement of the following analyzes. For the evaluation of cardiac function, the left ventricle was catheterized. Proteins expression in the left ventricle were performed by Western Blotting. Collagen and hypertrophy were assessed by histology and the citrate synthase activity by fluorimetry. After eight weeks of exercise training and myocardial infarction, animals of OVX+IMEF group showed a reduction in the expression of AT1 receptor and gp91phox, accompanied by an increase in catalase expression when compared to MI group (p<0.05). In addition, the exercise training prevented the increase in left ventricular end-diastolic pressure together with the reduction of dP/dt+. The cross-sectional area of the myocyte and collagen deposition were significantly increased in the MI group (p<0.05), but these changes were also prevented by exercise training. Together, these results demonstrates the beneficial effects promoted by the exercise training in treadmill on the cardiac remodeling process, impacting directly on the improvement of cardiac function in ovariectomized rats submitted to myocardial infarction

O remodelamento ventricular após um evento agudo isquêmico é o principal fator envolvido na disfunção ventricular e apresenta importantes diferenças entre machos e fêmeas. Estudos têm demonstrado que a redução dos hormônios ovarianos pode interferir de forma negativa em importantes vias relacionadas com o processo de remodelamento ventricular. Além disso, a prática regular de exercício físico tem demonstrado importantes efeitos benéficos após o infarto do miocárdio. Diante disso, os objetivos do presente estudo é avaliar os efeitos do treinamento físico de corrida em ratas ovariectomizadas após o infarto do miocárdio sobre a função cardíaca, parâmetros histológicos e morfométricos e expressão de proteínas relacionadas ao sistema renina angiotensina, sistema oxidante e antioxidante cardíaco. Foram utilizadas ratas da linhagem Wistar (Rattus Norvegicus Albinus), com oito semanas de idade, ovariectomizadas e divididas aleatoriamente em cinco grupos: Controle, OVX+SHAMSED, OVX+SHAMEF, OVX+IMSED e OVX+IMEF. Duas semanas após a indução do infarto do miocárdio os animais iniciaram o treinamento de corrida. O protocolo de treinamento foi realizado por um período de oito semanas, com duração de 60 min/dia, cinco vezes por semana com intervalo de dois dias para repouso. Quarenta e oito horas após o último dia do protocolo de treinamento físico os animais foram anestesiados e as seguintes análises realizadas: (a) avaliação da função cardíaca, após a cateterização do ventrículo esquerdo; (b) a avaliação da expressão proteica das enzimas antioxidantes (SOD e Catalase), do receptor AT1 de angiotensina II e da gp91phox no ventrículo esquerdo pela técnica de Western Blotting; (c) avaliação da deposição de colágeno e da hipertrofia do ventrículo esquerdo por morfometria; (e) a atividade da enzima citrato sintase no músculo sóleo por fluorimetria. Os resultados demonstram que após oito semanas de treinamento físico, os animais do grupo OVX+IMEF apresentaram uma redução na expressão do receptor AT1, acompanhado do aumento na expressão da Catalase quando comparado ao grupo OVX+IMSED (p<0,05). Além disso, o treinamento físico preveniu o aumento da pressão diastólica final do ventrículo esquerdo juntamente com o aumento do dP/dt+. A área de secção transversa do miócito e a deposição de colágeno foi significativamente maiores no grupo OVX+IMSED do que no grupo com OVX+IMEF (p<0,05).
