Biorremediação de azul de indigotina por lacase imobilizada em goma polissacarídeos purificados de Anadenanthera colubrina

Resumo: | Lacasses are capable of oxidizing various pollutants, suggesting their potential in the treatment of effluents with dyes. It was tried to produce beads with Anadenanthera colubrina gum and polysaccharides to immobilize the laccase and to use it for the bioremediation of indigotine blue dye. The extraction of polysaccharides from A. colubrina was optimized by factorial planning using as variables the pH, temperature and concentration of the gum. Pycnoporus sanguineus was used to produce laccase being the residue of the bark of Theobroma cacao as the substrate. The beads were optimized to immobilize the laccase produced by varying the concentration of the structural reagents. Immobilized laccase beads were used to bioremediate the indigotin blue dye 10 mg/L, compared to the use of the free enzyme. The optimized extraction condition of the polysaccharides were pH 5.75, temperature 30 ° C with yield of 83,01%. The laccase produced with the T. cacao residue had better enzymatic activity on day 6 of 3,000 U.mL-1. The beads optimized with the immobilized enzyme were the 16P formulation, therefore the purified polysaccharides were better used for the immobilization of the laccase in the beads. The laccase was efficient in bioremediation 94% of the dye, and the beads with immobilized enzyme bioremediated 68%. |

Procurou-se produzir “beads” com a goma e polissacarídeos da Anadenanthera colubrina para imobilização da lacase e utilizá-la para a biorremediação do corante azul de indigotina. Otimizou-se a extração dos polissacarídeos de A. colubrina por planejamento fatorial tendo como variáveis o pH, temperatura e concentração da goma. Utilizou-se Pycnoporus sanguineus para produzir lacase tendo como substrato resíduo da casca de Theobroma cacao. Otimizou-se a produção da “beads” para imobilização da lacase produzida variando a concentração dos reagentes estruturais. As “beads” com a lacase imobilizada foram utilizadas para biorremediar o corante azul de indigotina 10 mg/L, comparando com o uso da enzima livre. A condição otimizada de extração dos polissacarídeos foram pH 5,75, temperatura 30 ºC, com o rendimento de 83,01%. A lacase produzida com o resíduo de T. cacao teve melhor atividade enzimática no 6º dia de 3.000 U.mL-1. A “beads” otimizada com a enzima imobilizada foi a formulação 16P, sendo melhor utilizar os polissacarídeos purificados para a imobilização da lacase nas “beads”.
