Resumo: Sheep have been widely used as an experimental model for various anesthetic and surgical procedures, there is a shortage of information on the pharmacokinetics and hemodynamic behavior of anesthetic drugs in this species. No studies were found comparing the use of doppler echocardiography with a thermodilution in the measurement of the cardiac output of the animals. The objective of the study was to evaluate the pharmacological, hemodynamic and compare analyzes of the measurement of the cardiac output of sheep submitted to anesthesia with propofol. For this purpose, 10 healthy, uncastrated male sheep weighing 36 ± 4 kg on average were submitted to general anesthesia with propofol. One recommended induction dose to 6.5 mg / kg and the maintenance rate 0.5 mg / kg / minute. The protocol was determined by evaluating the loss of eyelid and laryngeal reflexes, allowing the accomplishment of endotracheal intubation. At time T0, T2, T5, T15, T30, T60, T65, T70, T75 were performed as collections of blood samples for pharmacokinetics through chromatography and evaluation of hemodynamic and respiratory parameters (HR, SBP, MAP, DAP). As well as measurements of cardiac output by Doppler echocardiography obtained at time T0, T5, T15, T30, T60. They were statistically evaluated by analysis of variance in a plot of subdivided plots without time, by Tukey test at 5% probability.The mean differences with the CO assessment methods were evaluated by the T test for paired data, along with an estimate of correlation between the data. A regression equation was also estimated to determine the values of the thermodilution technique as a function of the Doppler echocardiographic values. The evaluation of CO measurement methods, besides the comparison of the means obtained between the methods, evaluated the behavior of the values within the same technique over time. Plasma concentration of propofol showed a behavior in all animals during the evaluation period, generating its volume of concentration (56.2 ± 8.0 μg / mL) without non-T2 plasma (Tmax). The hemodynamic parameter was also observed as a reduction of blood pressure and PVR and of respiratory parameters and respiratory acidosis. It was observed that the CO values obtained by Doppler echocardiography remained always higher when compared to a Swan-Ganz catheter thermodilution. However, when the regression equation of the Doppler echocardiography values is applied as a function of thermodilution, very close values are obtained. Propofol induction without addition, but respiratory depression is identified by the increase in Paco2 values. Reduced blood pressure, as well as cardiac and systolic indexes are attributed to decreased peripheral vascular resistance. The Doppler echocardiographic method produces always higher values of CO when compared to those obtained by thermodilution, but it is possible to follow one direction of the changes in CO, in a reliable way, to use as two measurement techniques.

Não são encontrados estudos que compararam a utilização da ecodopplercardiografia com a termodiluição na mensuração do débito cardíaco desses animais. O objetivo do estudo foi avaliar as alterações, farmacocinéticas, hemodinâmicas e comparar duas técnicas de mensuração do débito cardíaco de ovinos submetidos a anestesia com propofol. Para tanto, 10 ovinos machos não castrados, saudáveis, com peso médio de 36±4kg foram submetidos à anestesia geral com propofol. A dose de indução preconizada foi de 6,5mg/kg e a taxa de manutenção 0,5mg/kg/minuto. O protocolo foi determinado avaliando a perda dos reflexos palpebrais e laríngeos permitindo realização de intubação endotraqueal. Nos tempos T0, T2, T5, T15, T30, T60, T65, T70, T75 foram realizadas as coletas das amostras de sangue para realização do perfil farmacocinético por meio da cromatografia líquida de alta eficiência e avaliação dos parâmetros hemodinâmicos e respiratórios (FC, PAS, PAM, PAD, RVP, PVC, DC, IC, VS, IS, ƒ, Etco2, Paco2 e pH), assim como as mensurações do débito cardíaco pela ecodopplercardiografia foram obtidos nos tempos T0, T5, T15, T30, T60. Foram avaliados estatisticamente pela análise de variância em esquema de parcelas subdivididas no tempo, pelo teste de Tukey à 5% de probabilidade. As diferenças médias entre os dois métodos de avaliação do DC foram avaliadas pelo teste T para dados pareados, juntamente com a estimativa de correlação entre os dados. Também foi estimada a equação de regressão, para determinar os valores do débito da técnica de termodiluição em função dos valores ecodopplercardiográficos. Para avaliação dos métodos de mensuração do DC, além da comparação das médias obtidas entre os métodos, avaliou-se o comportamento dos valores dentro da mesma técnica ao longo do tempo. A concentração plasmática do propofol apresentou um comportamento semelhante em todos animais durante o período de avaliação, atingindo seu maior nível de concentração (56,2±8,g/mL) no plasma no T2 (Tmax). Observou-se também alterações dos parâmetros hemodinâmicos como a redução da pressão arterial e da RVP e dos parâmetros respiratórios e acidose respiratória. Foi observado que os valores do DC obtidos por meio da ecodopplercardiografia se mantiveram sempre maiores quando comparados com a termodiluição com cateter de Swan-Ganz. Entretanto, quando se aplica a equação de regressão dos valores da ecodopplercardiografia em função da termodiluição são obtidos valores muito próximos a estes. O propofol proporciona indução sem apnéia, porém depressão respiratória é identificada pelo aumento dos valores da Paco2. Redução da pressão arterial, assim como dos índices cardíaco e sistólico são atribuídos a diminuição da resistência vascular periférica. As well as measurements of cardiac output by Doppler echocardiography obtained at time T0, T5, T15, T30, T60. They were statistically evaluated by analysis of variance in a plot of subdivided plots without time, by Tukey test at 5% probability.The mean differences with the CO assessment methods were evaluated by the T test for paired data, along with an estimate of correlation between the data. A regression equation was also estimated to determine the values of the thermodilution technique as a function of the Doppler echocardiographic values. The evaluation of CO measurement methods, besides the comparison of the means obtained between the methods, evaluated the behavior of the values within the same technique over time. Plasma concentration of propofol showed a behavior in all animals during the evaluation period, generating its volume of concentration (56.2 ± 8.0 μg / mL) without non-T2 plasma (Tmax). The hemodynamic parameter was also observed as a reduction of blood pressure and PVR and of respiratory parameters and respiratory acidosis. It was observed that the CO values obtained by Doppler echocardiography remained always higher when compared to a Swan-Ganz catheter thermodilution. However, when the regression equation of the Doppler echocardiography values is applied as a function of thermodilution, very close values are obtained. Propofol induction without addition, but respiratory depression is identified by the increase in Paco2 values. Reduced blood pressure, as well as cardiac and systolic indexes are attributed to decreased peripheral vascular resistance. The Doppler echocardiographic method produces always higher values of CO when compared to those obtained by thermodilution, but it is possible to follow one direction of the changes in CO, in a reliable way, to use as two measurement techniques.
