Caracterização e criopreservação de sêmen de tambaqui (Colossoma macropomum) em diferentes crioprotetores

Resumo: The objective of the study is to characterize the semen of tambaqui Colossoma macropomum during the breeding season and to evaluate the efficiency of four cryoprotectants added to the extender on sperm cryopreservation of tambaqui and to define the best protocol for thawing. Three semen samples were collected at regular intervals from ten males (pool). In the first stage of the experiment the characterization of semen in natura was performed and it was subjected to the routine analysis in optical microscopy: motility, vigor, life time and counting the number of spermatozoa. In the second stage of the experiment cryopreservation of semen was performed. The cryoprotectants glycerol, dimethylsulfoxide (DMSO), dimethylformamide (DMF) and methanol were tested at a ratio of 10.0%. For the assessment of viability and comparison of treatments testing of post-thaw samples in vivo was performed in three different protocols for each cryoprotectant, in thawing time 15, 30 and 45 seconds, 30°C temperature, totalizing 12 treatments with five replicates each. Quantitative variables were subjected to analysis of variance (ANOVA) and Tukey test while qualitative variables were subjected to the Freedman test with 0.05 significance. No significant differences (p> 0.05) were observed for most of the evaluated parameters. The sperm concentration was higher in the first sampling (14.54 billion sperm /mL) and the lifetime decreased at the end of the experiment. The cryoprotectant medium containing methanol and thawing for 30 s was the treatment that showed the best results fertilization (77.65%/).

The cryoprotectant medium containing methanol and thawing for 30 s was the treatment that showed the best results fertilization (77.65%/).Objetivou-se com o trabalho caracterizar o sêmen detambaqui durante a época reprodutiva e avaliar a eficiência de quatro crioprotetores adicionados ao meio diluidor na criopreservação do sêmen de tambaqui (Colossoma macropomum) e definir o melhor protocolo de descongelamento. Foram realizadas três coletas com intervalos regulares de amostras de sêmen (pool) de dez reprodutores. Na primeira etapa do experimento foi realizada a caracterização do sêmen in natura, sendo submetido a análises de rotina em microscopia óptica: motilidade, vigor espermático, tempo de vida e contagem do número de espermatozoides. Na segunda etapa do experimento foi realizada a criopreservação do sêmen. Os crioprotetores testados foram: glicerol, dimeltilsulfóxido (DMSO), dimetilformamida (DMF) e metanol, na proporção de 10,0%. Para a avaliação da viabilidade e comparação dos tratamentos foi realizado o teste in vivo das amostras pósdescongelamento em três protocolos diferentes para cada crioprotetor, em que foram testados os tempos de descongelamento de 15, 30 e 45 s, na temperatura de 30ºC, totalizando 12 tratamentos com cinco repetições. As variáveis quantitativas foram submetidas à análise de variância (ANOVA) e teste de Tukey e as variáveis qualitativas ao teste de Friedman, com significância de 0,05. Não foram observadas diferenças significativas (p>0,05) para a maioria dos parâmetros avaliados. A concentração espermática foi maior na primeira coleta (14,54 bilhões de espermatozoides/mL) e o tempo de vida diminuiu ao final do experimento.
