IN VITRO ACTIVITY OF SECONDARY METABOLITES OF Burkholderia sp. (BURKHOLDER 1950) AGAINST THE YEAST Malassezia pachydermatis (WEIDMANN) DODGE 1935.

Resumo: | M. pachydermatis is among the main agents involved in skin lesion and otopathological alterations of dogs and cats frequently associated to bacterial infections and host immune suppression. Because of resistance toward drugs, the market urge new therapies for treatment of fungal infections are needed. The effect of metabolites present in the filtrate culture of Burkholderia sp. (FCB) was tested against native clinical strains of the yeast M. pachydermatis in liquid medium measured by standard growth curve, by agar diffusion method, and colony forming units count in agar. Activity of FCB on cells was evaluated by transmission eletron microscopy (TEM). For comparison, sensitivity tests were performed on the yeast by diffusion agar test using commercial drugs. Crude FCB presented activity against the yeast by the methods utilized. The activity of FCB on cells was also observed by TEM, which revealed loss of eletrondensity and organelles structure alteration. These results suggest internal and external activities on the yeast cells and the potential use of FCB against M. pachydermatis. Novas terapias para o tratamento de infecções fúngicas são necessárias por causa do aparecimento de cepas resistentes. O efeito inibitório de metabólitos presentes no filtrado de cultivo da bactéria Burkholderia sp.(FCB) foi testado sobre cepas nativas clínicas da levedura M. pachydermatis em meio líquido pelo padrão de curva de crescimento, pela técnica de difusão em ágar e contagem de unidades formadoras de colônias em meio sólido. Avaliou-se a atividade de FCB sobre as células tratadas por microscopia eletrônica de transmissão (MET). Utilizaram-se antifúngicos comerciais para efeitos de comparação. FCB apresentou atividade sobre a levedura pelas técnicas utilizadas. Nas análises de MET, os efeitos de FCB sobre as células também foram observados por alterações da parede com diminuição da eletrondensidade e desestruturação das organelas. Esses resultados sugerem a atividade interna e externa sobre a levedura e o uso potencial de FCB contra M. pachydermatis.
