Resumo: | Siparuna guianenis Aublet is an aromatic plant, popularly known as negramina. Its leaves are traditionally used in the treatment of several diseases, as an anti-inflammatory and antipyretic. This work aims to verify the anatomical characteristics of the leaf blade, the toxicity, the anti- inflammatory potential, the capacity of caudal fin regeneration and activity of antioxidant enzymes of the aqueous (AE), hydroethanolic (HE), methanolic (ME) and essential oil (EO) extracts of S. guianensis leaves in zebrafish embryos and larvae. To verify the anatomical characteristics, an equation was determined to estimate the leaf area, the quantification and characterization of stomata and different histochemical methods were used. From the leaves, three types of extracts were prepared using different solvents (methanol, alcohol 70o and distilled water) and the EO was obtained by hydrodistillation. The main phenolic compounds present in the extracts were determined using HPLC analysis and the identification and quantification of the EO by chromatographic analysis. Toxicity and teratogenicity were investigated using the zebrafish model. Zebrafish embryos were treated with different concentrations of the extracts (10 to 0.08mg/ml) and OE (200 to 0.39μg/ml) for a period of 120 hours, and mortality, hatching and malformations were evaluated. The non-lethal and teratogenic concentrations were used for the analyses of neutrophil migration; caudal fin regeneration; apoptosis and activity of the antioxidant enzymes: superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST). It was identified in the anatomical analysis of the leaves the presence of stomas in the epidermis to classify the leaves as anfihypoestomatics. The existence of stellate tector trichomes in the abaxial epidermis and the presence of secondary metabolites such as phenolic compounds, alkaloids, tannin and lignin were identified and the mathematical equation obtained was through linear regression by means of the length and width relationship, using the model Y=3.5833+0.6715x, with a determination coefficient of 0.97. It was identified in the extracts the presence of phenolic compounds with trans-cinnamic acid. The extracts showed toxicity, being for EA at concentrations from 2.5 to 10mg/ml and for EH and EM between 0.62 to 10mg/ml. The larvae treated with EA didn’t exhibit malformations. All tested extracts demonstrate anti-inflammatory activity, however they do not show good performance in caudal fin regeneration. The extracts did not influence SOD and CAT activity. EA didn’t induce apoptotic cells unlike EH and EM. In OE was identified the presence of the monoterpene β-Myrcene (24.72%). In the inflammation test, the different concentrations showed ability to reduce the migration of neutrophils to the site of the larvae, in the regeneration of the caudal fin the EA showed a lower regeneration rate than the controls (DMSO 1% and Medium E3). No influence of EO on antioxidant enzymes was verified. Finally, apoptotic cells were identified in the region of the heart at the concentration of 12.5μg/ml. Conclude that the stomata of this species are classified as paracytic and the EA, EH, EM and the OE of the leaves of S. guianensis present anti-inflammatory activity by reducing the migration of neutrophils. |

Suas folhas são utilizadas tradicionalmente no tratamento de diversas enfermidades, como anti- inflamatório e antipirético. Este trabalho tem como objetivo verificar as características anatômicas da lâmina foliar, a toxicidade, o potencial anti-inflamatório, a capacidade de regeneração da nadadeira e atividade das enzimas antioxidantes dos extratos aquoso (EA), hidroetanólico (EH), metanólico (EM) e do óleo essencial (OE) das folhas S. guianensis em embriões e larvas de zebrafish. Para verificar as características anatômicas foi realizada à determinação de uma equação para estimar a área foliar, a quantificação e caracterização dos estômatos e foram utilizados diferentes métodos histoquímicas. A partir das folhas foram elaborados três tipos de extratos empregados diferentes solventes sendo, metanol, álcool 70o e água destilada; e a obtenção do OE foi realizada por hidrodestilação. Os principais compostos fenólicos presentes nos extratos foram determinados usando análise HPLC e a identificação e quantificação do OE através da análise cromatográfica. A toxicidade e teratogenicidade foram investigadas usando o modelo zebrafish. Embriões de zebrafish foram tratados com diferentes concentrações dos extratos (10 a 0,08mg/ml) e OE (200 a 0,39μg/ml) durante um período de 120 horas, sendo avaliadas mortalidade, eclosão e malformações. As concentrações não letais e teratogênicas foram utilizadas para a realização das análises de migração de neutrófilos; regeneração da nadadeira; apoptose e atividade das enzimas antioxidantes: superóxido dismutase (SOD), catalase (CAT) e glutationa-S-transferase (GST). Foi identificada na análise anatômica das folhas a presença de estômatos na epiderme classificar as folhas como anfihipoestomática. Foi identificada a existência de tricomas do tipo tectores estrelados na epiderme abaxial e a presença de metabólitos secundários, como compostos fenólicos, alcaloide, tanino e lignina, e a equação matemática obtida foi através da regressão linear por meio da relação comprimento e largura, utilizado o modelo Y=3,5833+0,6715x, com coeficiente de determinação de 0,97. Foi identificada nos extratos a presença de compostos fenólicos com o ácido trans-cinâmico. Os extratos apresentaram toxicidade, sendo para o EA nas concentrações de 2,5 a 10mg/ml e os EH e EM entre 0,62 a 10mg/ml. As larvas tratadas com EA não exibiram malformações. Todos os extratos testados demonstram atividade anti- inflamatória, no entanto não demonstram bom desempenho na regeneração da nadadeira. Os extratos não influenciaram na atividade da SOD e CAT. EA não induziu células apoptóticas ao contrário dos EH e EM. No OE foi identificado à presença do monoterpeno β-Micerno (24,72%). No teste de inflamação, as diferentes concentrações apresentaram capacidade de reduz à migração dos neutrófilos para o local lecionado das larvas, na regeneração da nadadeira caudal o OE demonstrou uma taxa de regeneração menor que os controles (DMSO 1% e Meio E3). Não foi verificada a influência do OE nas enzimas antioxidantes, por fim foram identificadas células apoptóticas na região do coração, na concentração de 12,5μg/ml.
