Resumo: Lettuce is one of the vegetables with the highest expression in the world. Its consumption is growing every year in Brazil and expanded the production areas for different regions. However, a limitation for its production is the occurrence of high temperatures during the germination, which takes most cultivars do not germinate at temperatures above 28 ° C. However, the Everglades cultivar shows germination of 60 to 70%, on average 35ºC, considered tolerant germination at high temperature (tolerant thermo inhibition).. On the other hand, also a factor that normally prevents / reduces germination soon after harvesting is the primary dormancy. The position of the seed in the mother plant is an aspect that can influence the dormancy of lettuce seeds. Thus, the aim of this work was to evaluate the influence of the position of the seed in the mother plant and the effect of Everglades cultivar lettuce seed germination in storage at 35 ° C temperature, and analyze the levels of enzymes and proteins heat tolerant. The experiments were conducted in the experimental area of HortiAgro Sementes S/A and Seed Analysis Laboratory (UFLA). Were obtained progenies of Everglades cultivars (tolerant thermoinhibition) and Veronica (sensitive to thermoinhibition) from germinated seeds in temperatures of 20 and 35 ° C, the seeds, object of study, collected in two parts of the inflorescence (apex and base). The seeds of the progenies were evaluated by germination in four seasons after harvest (70, 130, 190 and 400 days), these were also evaluated activity of endo-β-mannanase enzyme and standard resistant protein to heat to 400 days after the harvest. There was no difference in seed germination as to its origin in the previous generation, both in plants grown from seeds that germinated at 20 or 35 ° C. Was also not found difference in the position of the seed in the mother plant inflorescences. On the other hand, there were differences between the progenies of "Everglades" and the storage times. The Everglades cultivar showed to be tolerant thermoinhibition compared to cultivate Veronica and that this tolerance is affected by the primary dormancy / thermoinhibition. Overall there was no direct relationship between the progenies, the percentage of germination and enzyme activity. It was also found that the expression of the protein and the activity of endo-β-mannanase is higher in the thermoinhibition tolerant genotype.

Seu consumo cresce a cada ano em todo o Brasil e ampliam-se as áreas de produção para diferentes regiões. Porém, uma das limitações para sua produção é a ocorrência de temperaturas elevadas durante a germinação das sementes, o que leva a maioria das cultivares a não germinarem em temperaturas superiores a 28 °C. No entanto, a cultivar Everglades apresenta germinação de 60 a 70%, em média, em temperatura de 35 ºC, sendo considerada tolerante à germinação em temperatura alta (tolerante à termoinibição). Por outro lado, ainda, um fator que normalmente impede/diminui a germinação logo após a colheita é a dormência primária. A posição das sementes na planta-mãe é um aspecto que pode influenciar a dormência de sementes de alface. Assim, objetivou-se com este trabalho, avaliar a influência da posição das sementes na planta-mãe e o efeito do armazenamento de sementes de alface da cultivar Everglades na germinação em temperatura de 35 ºC, e analisar a atividade de enzimas e bandas de proteínas tolerantes ao calor. Os experimentos foram conduzidos na área experimental da HortiAgro Sementes S/A e no Laboratório de Análises de Sementes (UFLA). Foram obtidas progênies das cultivares Everglades (tolerante à termoinibição) e Verônica (sensível à termoinibição) a partir de sementes germinadas nas temperaturas de 20 e 35 °C, sendo as sementes, objeto de estudo, colhidas em duas partes da inflorescência (ápice e base). As sementes das progênies foram avaliadas pelos testes de germinação em 4 épocas após a colheita (70, 130, 190 e 400 dias), nestas também foram avaliadas atividade da enzima endo-β-mananase e padrão da proteína resistente ao calor aos 400 dias após a colheita. Não houve diferença na germinação das sementes quanto à sua origem na geração anterior, seja de plantas oriundas de sementes que germinaram a 20 ou a 35 ºC. Também não se constatou diferença quanto à posição das sementes nas inflorescências da planta mãe. Por outro lado, verificaram-se diferenças entre as progênies de “Everglades” e para as épocas de armazenamento. A cultivar Everglades mostrou-se tolerante à termoinibição quando comparada à cultivar Verônica e sendo essa tolerância afetada pela dormência primária/termoinibição. De modo geral não houve relação direta entre as progênies, a percentagem de germinação e a atividade enzimática.
