Multiplicação in vitro do porta-enxerto de videira 'VR043-43' (Vitis vinifera x Vitis rotundifolia)

Resumo: The micropropagation is one technique that makes possible the massal propagation of plants of economic interest. The objective of this work was to evaluate the effect of 6–benzilaminopurina (BAP) and kinetin (KIN) in the multiplication in vitro of the grapevine rootstock 'VR043–43'. The isolation was carried through in culture medium QL, supplemented with BAP or KIN, in the concentrations of 0; 2,5; 5,0 and 10,0µM. The subcultures had been carried out each 45 days. The number of shoots per explant increased along with the subcultures with BAP. However this effect was not observed with the KIN that it got resulted similar to the one of the cytokin–free (1,0 shoot/explant). The concentrations of 5,0 nd 10,0 µM of BAP had promoted the biggest number of shoots with reduction of the height. The highest concentration of KIN (10,0 µM) also had negative effect in the height of the shoots, as well as, in the number of leaves per shoot. The callus formation (100%) was observed with BAP and KIN in all the concentrations, and in the cytokinin absence it did not have the callus formation. Cytokinin BAP reduced the formation of roots that was of 100% with the KIN and cytokinin–free.

Objetivou-se com este trabalho avaliar o efeito de 6-benzilaminopurina (BAP) e cinetina (CIN) na multiplicação do porta-enxerto de videira 'VR043-43'. O isolamento foi realizado em meio de cultura QL, suplementado com BAP ou CIN, nas concentrações de 0; 2,5; 5,0 e 10,0µM. Os subcultivos foram realizados a cada 45 dias. O número de brotações por explante aumentou ao longo dos subcultivos com BAP. Esse efeito não foi observado com a CIN, que obteve resultado semelhante ao da testemunha (1,0 brotação/explante). As concentrações de 5,0 e 10,0 µM de BAP promoveram o maior número de brotações, porém com redução da altura. A concentração mais elevada de CIN (10,0 µM), também teve efeito negativo na altura das brotações, assim como no número de folhas por brotação. A formação de calo (100%) foi observada com BAP e CIN em todas as concentrações, e na ausência de citocinina não houve a formação de calo.
