Resumo: Osteoarthritis is a worldwide degenerative chronic disease. Approximately 250 million people perform worldwide. Thus, the new forms of treatment are the disease of fundamental importance. The objective of this study was to evaluate the hydroethanolic extraction of leaves of Persea americana Mill in an experimental model of osteoarthritis in rat knees. Chemical characterization of the extract using phytochemical screening and high efficiency HLPC-UV array ethnography at 254 nm, identification by Mass Spectrometry Coupled Liquid Chromatography (LC-MS) and fluid flow analysis (FIAMS). Use and control of Wistar male males divided into two groups: EHPA (500mg / kg), saline (0.1mL) / kg), meloxicam (0.1mg / kg) and control subgroup that had no intervention.) For evaluation for motor activity the forced ambulation test was used. This was to quantify the frequency the Von-Frey test was used. A response to mechanical hyperalgesia was evaluated by the Randall Selitto test. Biochemical markers such as fasting glucose, total cholesterol, triglycerides, creatinine, urea, gamma GT, oxalacetic transaminase (AST) and pyruvic transaminase (ALT) were performed. They were also histological retrospectives of the knee joint using the OARSI criteria and radiographic analysis following the Kellgren and Lawrence osteoarthritis grade parameters. Qualitative phytochemical screening shows the presence of phenolic compounds, alkaloids, triterpenoid steroids, tannins and saponins. A chemical characterization by LC-MS and FIA-MSH. A presence of condensed tannins as major components, as well as phenolic acids, aglycone, glycosylated and triterpene flavonoids. Oral administration of the extract at a dose of 500mg / kg improved the dose of 500mg / kg orally and improved antinociceptive activity in the mechanical hyperalgesia test in both experiments. The extract did not alter liver, renal and hematopoietic functions. The histological and radiological analyzes that were extracted were a protector of cartilage in the combat process. Concluding the hydroethanolic extract of Persea americana Mill. reduced sensitivity to blood damage to adverse effects of meloxicam, a drug commonly used in medical practice.

Aproximadamente 250 milhões de pessoas apresentam osteoartrite de joelho em todo o mundo. Dessa forma, encontrar novas formas de tratamento dessa doença é de fundamental importância. O estudo teve por objetivo avaliar os efeitos do extrato hidroetanólico das folhas de Persea americana Mill. em modelo experimental de osteoartrite em joelhos de ratos. Para caracterização química do extrato utilizou-se screening fitoquímico e perfil cromatográfico obtido por HLPC-UV de alta eficiência a 254 nm, identificação por Cromatografia Líquida Acoplada à Espectrometria de Massas (LC-MS) e análise da injeção de fluxo (FIA-MS). Utilizou-se quarenta e oito ratos machos da linhagem Wistar divididos em dois grupos (tratamento imediato e tardio), onde cada um desses dois grupos foram divididos em quatro subgrupos com seis animais: EHPA (500mg/kg), solução salina (0,1mL/kg), meloxicam (0,1mg/kg) e subgrupo sadio que não possuiu intervenção cirúrgica e tratamento. Para avaliação de atividade motora foi utilizado o teste de deambulação forçada, para quantificar a dor espontânea foi utilizado o teste de Von-Frey, e a resposta frente a hiperalgesia mecânica foi avaliada pelo teste de Randall Selitto. Realizou-se dosagens de marcadores bioquímicos, tais como glicemia em jejum, colesterol total, triglicerideos, Creatinina, Ureia, Gama GT, Transaminase Oxalacetica (AST) e Transaminase Piruvica (ALT), além do hemograma completo. Também foram realizadas análises histológicas da articulação do joelho utilizando os critérios da OARSI e análise radiográfica seguindo os parâmetros de grau osteoartrose de Kellgren e Lawrence. O screening fitoquímico qualitativo mostrou a presença de compostos fenólicos, alcaloides, esteroides triterpenóides, taninos e saponinas. A caracterização química por LC-MS e FIA-MS revelou a presença de taninos condensados como componentes majoritários, além de acidos fenólicos, flavonoides agliconas, glicosilados e triterpenos. A administração do extrato na dose de 500mg/kg, via oral, melhorou a deambulação forçada, reduziu a sensibilidade tátil e mostrou atividade antinociceptiva no teste de hiperalgesia mecânica em ambos protocolos experimentais. O extrato não alterou as funções hepáticas, renais e hematopoiéticas. As análises histológicas e radiológicas mostraram que o extrato atuou como um protetor da cartilagem no processo inflamatório.
