Apesar disso, os asininos quando infectados são capazes de suprimir os sinais clínicos e manter o título viral abaixo do limiar de provocar doença ao longo de todo período de infecção. Aspectos associados à resposta imune humoral e celular podem estar envolvidos no quadro clínico diferente encontrado em jumentos infectados com o Equine infetious anemia virus (EIAV), dentre eles, os fatores de restrição retrovirais (RRFs), proteínas induzíveis por interferons, que limitam alguma etapa do ciclo de replicação viral e a expressão diferenciada do receptor de lentivírus equino tipo I (ELR1) que está associado à entrada do vírus na célula-alvo. Nesse sentido, esse trabalho propôs analisar a expressão dos RRFs APOBEC3, ADAR1, RSAD2, SAMHD1, Mx2, BST2, SERINC e TRIM5α, bem como comparar a expressão do ELR1 na sua forma funcional e a presença de isoformas solúveis em células mononucleares de cavalos e jumentos infectadas com o EIAV, in vitro. Para isso, o sangue total de 3 cavalos e 3 jumentos foram submetidos ao isolamento e cultivo das células mononucleares do sangue periférico (PBMC), seguindo de infecção com a estirpe patogênica EIAVWyo. Nossos resultados demonstraram que o ADAR1 é mais expresso nos equinos, assim como o Mx2. Já os RRFs A3Z3, TRIM5α e SERINC, são mais expressos nos macrófagos de jumentos quando comparado aos de cavalos. Com relação ao ELR-1, foi observado que macrófagos de jumentos e de cavalos expressam igualmente o receptor ao longo do tempo, porém a isoforma solúvel do receptor (sELR1) é inicialmente mais expressa em macrófagos derivados de monócitos (MDM) de jumentos. Dessa forma, sugerimos que a expressão diferenciada dos RRFs e do sELR1 em MDM de jumentos quando comparado aos de cavalos, pode estar associado ao controle rigoroso da replicação do EIAV nessa espécie.Equine Infectious Anemia (EIA) is a viral disease that affects all members of the Equidae family. Despite this, donkeys, when infected, are able to suppress clinical signs and keep the viral titer below the threshold of causing disease throughout the entire period of infection. Factors associated with the humoral and cellular immune response may be involved in this different issue found in donkeys infected with the Equine infectious anemia virus (EIAV), including retroviral restriction factors (RRFs), interferon-inducible proteins that limit some stage of the viral replication cycle and the differentiated expression of the equine lentivirus receptor type I (ELR1) which is associated with virus entry into the target cell. In this sense, this work proposed to analyze the expression of the RRFs APOBEC3, ADAR1, RSAD2, SAMHD1, Mx2, BST2, SERINC and TRIM5α, as well as to compare the expression of ELR1 in its functional form and the presence of soluble isoforms in mononuclear cells of horses and donkeys infected with EIAV, in vitro. For this, whole blood from 3 horses and 3 donkeys were subjected to isolation and culture of peripheral blood mononuclear cells (PBMC), followed by infection with the pathogenic strain EIAVWyo. Our results demonstrated that ADAR1 is more expressed in horses, as well as Mx2. The RRFs A3Z3, TRIM5α and SERINC are more expressed in donkey macrophages when compared to horses. With regard to ELR-1, it was observed that macrophages from donkeys and horses equally express the receptor over time, however the soluble receptor isoform (sELR1) is initially more expressed in macrophages derived from monocytes (MDM) from donkeys.
