Efeitos da farinha de Pereskia aculeata Mill. (Ora-pro-nobis) no perfil corporal, bioquímico e molecular nos tecidos adiposos branco e marrom de camundongos submetidos a treino resistido

Resumo: Physical exercise is an important health promotion strategy, especially due to the modulation it exerts on various organs and tissues. Exercise performance can be optimized by different strategies, especially food. So, identification of dietary strategies with a focus on improving the performance of physical exercise allowing the increasing its effects is a promising field of investigation. Pereskia aculeata Mill. (ora pro-nobis) is a perennial species of the Cactaceae family, used mainly as a vegetable unconventional hardwood. Although it has great productive and nutritional potential, in addition to medicinal properties is still underexplored and little known. The present study aims to objective to evaluate the effects of ora-pro-nobis flour on body composition, biochemical and molecular expression of genes associated with lipolysis/lipogenesis in tissue white adipose tissue and thermogenesis in brown adipose tissue of mice submitted to resistance training. The study was conducted with 14 mice were randomized into two groups: G1: control (CONTROL), fed with standard diet + resistance training; G2: (OPN), fed with a modified standard diet and added ora-pro nobis flour + resistance training. The diet was produced from ora-pro-nobis flour. the flour was produced using leaves of the plant obtained in the region of Montes Claros, Minas Gerais, Brazil, which were washed, dried, dehydrated and crushed. The ora-pro-nobis flour diet presented final composition of 59.23% of carbohydrates, 28.07% of proteins and 12.7% of lipids, with an energy content of 3.03 Kcal/gram. The animals were submitted to 24 sessions of resistance training, composed of 6 sets (sets) of 8 repetitions with intervals of 120 seconds between sets. From the 13th section onwards, a weight was added to the training. attached to the animal's tail, corresponding to 50% of its animal's body weight. In the study body, biochemical and histological parameters were measured, in addition to the expression of markers associated with thermogenesis in brown adipose tissue and adipogenesis/lipolysis in white adipose tissue by the qRT-PCR technique. The results showed that the animals supplemented with oro-pro-nobis flour had lower mean weight values (Control: 48.03±1.06 vs. OPN: 45.0±0.53), weight gain (Control: 6.11±1.23 vs. OPN:9.95±1.03) and visceral body adiposity (Control: 0.031±0.002 vs. OPN: 0.024±0.002); in addition, they had higher energy expenditure and lipid oxidation rate and had lower LDL-c levels. In the molecular evaluation, they showed a decrease in mRNA expression. for ACC and increase for PCG1α in white and brown adipose tissue, respectively. It is concluded that orapro-nobis flour can modulate the effects of physical exercise in mice, improving body, biochemical and molecular parameters.

A performance do exercício físico pode ser otimizada por diversas estratégias, sobretudo alimentares. Assim, identificação de estratégias alimentares como foco na melhora da performance do exercício físico permitindo o incremento de seus efeitos é um promissor campo de investigação. Pereskia aculeata Mill. (orapro-nobis) é uma espécie perene da família Cactaceae, usada, principalmente, como hortaliça folhosa não convencional. Embora apresente grande potencial produtivo e nutricional, além de propriedades medicinais é ainda subexplorada e pouco conhecida. O presente estudo tem como objetivo avaliar os efeitos da farinha de ora-pro-nobis na composição corporal, parâmetros bioquímicos e na expressão molecular de genes associados à lipólise/lipogênese no tecido adiposo branco e da termogênese no tecido adiposo marrom de camundongos submetidos a treino resistido. O estudo foi conduzido com 14 camundongos foram randomizadas em dois grupos: G1: controle (CONTROL), alimentado com dieta padrão + treino resistido; G2: experimental (OPN), alimentado com dieta padrão modificada e adicionada farinha de ora-pronobis + treino resistido. A dieta foi produzida a partir da farinha de ora-pro-nobis. A farinha foi produzida com uso de folhas da planta obtidas na região de Montes Claros, Minas Gerais, Brasil, que foram lavadas, secadas, desidratadas e moídas. A dieta de farinha de ora-pro-nobis apresentou composição final de 59,23% de carboidratos, 28,07% de proteínas e 12,7% de lipídeos, com teor energético de 3,03 Kcal/grama. Os animais foram submetidos a 24 sessões de treino resistido, compostas por 6 conjuntos (séries) de 8 repetições com intervalos de 120 segundos entre os conjuntos. A partir da 13ª seção, foi incrementado ao treinamento, um peso fixado à cauda do animal, correspondente a 50% do seu peso corporal do animal. No estudo foram mensurados parâmetros corporais, bioquímicos, histológicos, além da expressão de marcadores associados à termogênese no tecido adiposo marrom e adipogênese/lipólise no tecido adiposo branco, pela técnica de qRT-PCR. Os resultados mostraram que os animais suplementados com farinha de oro-pro-nobis apresentaram menores valores de peso médio (Control: 48,03±1,06 vs. OPN: 45,0±0,53), do ganho de peso (Control: 6,11±1,23 vs. OPN: 9,95±1,03) e da adiposidade corporal visceral (Control: 0,031±0,002 vs. OPN: 0,024±0,002); ademais, tiveram maior gasto energético e taxa de oxidação de lipídeos e apresentaram menores níveis de LDL-c. Na avaliação molecular, apresentaram diminuição da expressão de mRNA para ACC e aumento para PGC1α no tecido adiposo branco e marrom, respectivamente.
