Trata-se de um problema de saúde pública que tem crescido no Brasil e no mundo em todos os estratos da sociedade. Muitos dos efeitos da obesidade sobre o desenvolvimento de distúrbios metabólicos e cardiovasculares já são conhecidos, no entanto, pouco se sabe sobre o impacto da obesidade sobre o sistema imunológico e as doenças infecciosas. Estudos recentes têm demonstrado que indivíduos obesos apresentam uma resposta imunológica diminuída predispondo ao aumento da susceptibilidade à infecção por diferentes agentes patogênicos. Assim sendo, o objetivo desse trabalho foi avaliar se a obesidade induzida por dieta hipercalórica altera a resposta imunológica em camundongos C57BL/6 frente à infecção por Leishmania major. Camundongos C57BL/6 foram divididos nos grupos AIN 93G (dieta normal) e HSF (dieta hipercalórica). Após 4 semanas de dieta os grupos foram infectados com L. major e eutanasiados 2, 4 e 8 semanas após a infecção. Avaliouse o ganho de peso, glicemia, lipídeos séricos, tamanho da lesão, parasitismo, perfil de citocinas e imunoglobulinas e produção de óxido nítrico. Os dados apresentados no presente estudo sugerem que a obesidade e suas complicações metabólicas parecem alterar a resposta imunológica durante à infecção por L. major. A alteração do perfil de citocinas (redução de IFN-γ e aumento de IL-10 ambos na segunda semana de infecção) possivelmente podem explicar os mecanismos envolvidos pelos quais os animais obesos apresentam maior tamanho de lesão e maior permissividade ao parasito.Obesity is a multifactor chronic disease and is associated with increased mortality, as it predisposes to a variety of diseases. This is a public health problem that has grown in Brazil and worldwide in all segments of society. Many effects of obesity on the development of metabolic and cardiovascular disorders are known, however little is known about the impact of obesity on the immune system and infectious diseases. Recent studies have shown that obese subjects have an impaired immune response predisposing to increased susceptibility to infection by various pathogens. Therefore, the aim of this study was to evaluate whether obesity induced by high calorie diet alters the immune response in C57BL / 6 mice to infection by Leishmania major. C57BL / 6 mice were divided into groups AIN 93G (normal diet) and HSF (hypercaloric diet). After 4 weeks of diet groups were infected with L. major and euthanized 2, 4 and 8 weeks after infection. We evaluated weight gain, blood glucose, serum lipids, lesion size, parasitism, cytokine profile and immunoglobulins and nitric oxide production. The data presented in this study suggest that obesity and its metabolic complications appear to alter the immune response during infection with L. major.
