O maropitant é um antiemético antagonista dos receptores NK-1 ligados a substância P, receptores estes que estão localizados no centro do vômito e também nas vias nociceptivas, sendo identificados em vísceras, medula espinhal e cérebro. O presente estudo teve como objetivo comparar o efeito antinociceptivo do maropitant em diferentes doses, em cadelas anestesiadas com sevoflurano submetidas à ovariohisterectomia por meio de parâmetros cardiorrespiratórios, ETSEVO e temperatura retal, além de avaliar o requerimento analgésico no pós-operatório por meio da Escala Composta de Dor de Glasgow (ECDG). Quarenta e cinco cadelas adultas, distribuídas em cinco grupos de nove animais, pesando em média 12,1 ± 4,2 kg foram sedadas com acepromazina (0,02 mg/kg, IM) e morfina (0,5 mg/kg, IM) para preparo cirúrgico. Após a indução com propofol dose resposta (1 a 6 mg/kg, IV) e manutenção do plano anestésico com sevoflurano, foi administrado, por via intravenosa, um bolus de maropitant (1 mg/kg), seguido de infusão contínua em doses crescentes em quatro grupos, sendo eles G50 (50 mcg/kg/h), G75 (75 mcg/kg/h), G100 (100 mcg/kg/h) e G200 (200 mcg/kg/h) e um grupo controle (n=9) que recebeu o volume equivalente da IC em NaCl 0,9%. Os parâmetros cardiorrespiratórios (FC, f, PAS/ PAD/ PAMinvasiva, SpO2, EtCO2), ETSEVO e temperatura retal foram avaliados em sete momentos distintos, sendo eles pré-indução (M0), pós-indução (M1), incisão cirúrgica (M2), tração do pedículo ovariano direito (M3), tração do pedículo ovariano esquerdo (M4), tração uterina (M5) e finalização da sutura cutânea (M6). Caso houvesse aumento de 20% nos valores de FC e PAM com relação ao M0, era realizado resgate analgésico com fentanil (1 mcg/kg, IV). Os escores de dor (ECDG) foram avaliados 1h, 2h, 4h e 6h após a cirurgia e foi administrado resgate com tramadol (5 mg/kg, SC) em caso de ECDG > 6. Para a análise dos dados foi realizada ANOVA seguida pelo teste F. As variáveis paramétricas foram comparadas por meio do teste de Tukey e as não-paramétricas pelos testes de Kruskal-Wallis e Skillings-Mack ou Friedman (p < 0,05). Não foram observadas diferenças significativas entre os protocolos analgésicos para as variáveis cardiorrespiratórias e temperatura retal. Quanto à ETSEVO, houve diferença estatística entre o GC e o G50 (p = 0,04) e entre o GC e o G100 (p = 0,03). Na avaliação da dor pós-operatória pela ECDG, não foi observada diferença significativa entre os grupos. Conclui-se que a administração do maropitant em infusão contínua no período transoperatório de cadelas submetidas à ovariohisterectomia demonstrou eficácia na redução do requerimento anestésico médio de sevoflurano nos grupos G50 e G100, no entanto, não foi suficiente para suprimir as respostas autonômicas aos estímulos nociceptivos somático e visceral e não demonstrou efeito evidente nos escores da avaliação da dor pós-operatória pela ECDG.Multimodal analgesia consists of the use of several drugs that have different mechanisms of action, aiming to promote adequate analgesia with a lower incidence of adverse effects. The present study aimed to compare the antinociceptive effect of maropitant in different doses, in bitches anesthetized with sevoflurane submitted to ovariohysterectomy by means of cardiorespiratory parameters, ETSEVO and body temperature, in addition to evaluate the analgesic requirement in the postoperative period using the Glasgow Pain Scale (ECDG). Forty-five adult female dogs, distributed in five groups of nine animals, weighing an average of 12.1 ± 4.2 kg were sedated with acepromazine (0.02 mg/kg, IM) and morphine (0.5 mg/kg, IM) for surgical preparation. After induction with propofol dose response (1 to 6 mg/kg IV) and maintenance of the anesthetic plan with sevoflurane, a bolus of maropitant (1 mg/kg) was administered intravenously, followed by continuous infusion of maropitant in increasing doses in four groups, they being G50 (50 mcg/kg/h), G75 (75 mcg/kg/h), G100 (100 mcg/kg/h) and G200 (200 mcg/kg/h) and a control group (n=9) that received the equivalent volume of CI in 0.9% NaCl. The cardiorespiratory parameters (HR, f, PAS / PAD / PAMinvasiva, SpO2, EtCO2), ETSEVO and rectal temperature were evaluated at seven different times, being preinduction (M0), post-induction (M1), surgical incision (M2 ), traction of the right ovarian pedicle (M3), traction of the left ovarian pedicle (M4), uterine traction (M5) and completion of skin suture (M6). If there was a 20% increase in HR and MAP values in relation to M0, analgesic rescue with fentanyl (1 mcg/kg, IV) was performed. Pain scores (ECDG) were assessed 1h, 2h, 4h and 6h after surgery and rescue with tramadol (5 mg/kg, SC) was administered in case of ECDG> 6. For data analysis, ANOVA was performed followed by the F test. Parametric measurements were compared using the Tukey test and non-parametric ones using the Kruskal-Wallis and SkillingsMack or Friedman tests with p <0.05. No significant difference was observed between analgesic protocols for cardiorespiratory variables and rectal temperature. As for ETSEVO, there was a statistical difference between the CG and the G50 (p = 0.04) and between the CG and the G100 (p = 0.03). In the assessment of postoperative pain by ECDG, no significant difference was observed between groups.
