A infecção é causada por Brucella canis, bactéria Gram-negativo que resulta em sinais clínicos inespecíficos nos cães e doença crônica debilitante em humanos. A vacinação é principal ferramenta para o controle da brucelose, no entanto, atualmente não existe vacina comercial disponível para o controle da brucelose canina. Este estudo teve como objetivo avaliar a proteção e a resposta imunológica induzida por Brucella ovis ∆abcBA (Bo∆abcBA) encapsulada com alginato contra o desafio por B. canis em camundongos e avaliar a segurança dessa cepa em cães. A cinética intracelular em macrófagos primários caninos e ovinos foi similar, sendo que as cepas selvagens B. ovis e B. canis foram capazes de multiplicar no meio intracelular, enquanto a cepa vacinal Bo∆abcBA apresentou perfil atenuado nestas células. A imunização de camundongos BALB/C com Bo∆abcBA (108 UFC – unidades formadoras de colônia) encapsulada em alginato induziu proliferação de linfócitos, produção de IL-10 e IFN-γ e protegeu contra o desafio experimental com B. canis. Cães imunizados com Bo∆abcBA (109 UFC) encapsulada em alginato soroconverteram e não apresentaram alterações hematológicas, bioquímicas ou clínicas. Além disso, Bo∆abcBA não foi detectada por isolamento ou PCR em amostras de sangue, sêmen, urina ou swab vaginal em nenhum momento no decorrer deste estudo. Bo∆abcBA foi isolado a partir de linfonodos próximos ao local da inoculação em dois cães 22 semanas após a imunização. Desta forma, os resultados mostram que Bo∆abcBA encapsulada em alginato protege camundongos contra o desafio experimental por B. canis, e é segura para cães.Canine brucellosis is an infectious and zoonotic disease with worldwide distribution and is considered neglected. The infection is caused by Brucella canis, a Gram-negative bacterium that results in unspecific clinical signs in dogs and chronic disease in humans. Vaccination is most important tool for controlling brucellosis, but currently there is no vaccine available for canine brucellosis. This study aimed to evaluate protection and immune response induced by Brucella ovis ∆abcBA (Bo∆abcBA) encapsulated with alginate against the challenge with B. canis in mice and to assess the safety of this strain for dogs. The intracellular kinetics in primary canine and sheep macrophages was similar, with the wild strains B. ovis and B. canis being able to multiply in the intracellular medium, while the Bo∆abcBA vaccine strain showed an attenuated profile in these cells. Immunization of BALB/C mice with alginate-encapsulated Bo∆abcBA (108 CFU – colony forming units) induced lymphocyte proliferation, production of IL10 and IFN-γ, and protected against experimental challenge with B. canis. Dogs immunized with alginate-encapsulated Bo∆abcBA (109 CFU) seroconverted, and had no hematologic, biochemic or clinical changes. Furthermore, Bo∆abcBA was not detected by isolation or PCR with blood, semen, urine samples or vaginal swabs at any time point over the course of this study. Bo∆abcBA was isolated from lymph nodes near to the site of inoculation in two dogs at 22 weeks post immunization.
