Com o objetivo de caracterizar as lesões intestinais nos diferentes segmentos e idade de aves acometidas, além de investigar a etiologia da RSS, pintinhos naturalmente infectados apresentando atraso no crescimento, apatia, alimento mal digerido nas fezes ou diarreia foram coletados de quatro diferentes granjas comerciais localizadas em três municípios do estado de Minas Gerais. Pintinhos sem sinais clínicos foram usados como controle. O número total de pintinhos examinados foi 243. A média de peso dos pintinhos acometidos foi menor que o esperado para os lotes. Macroscopicamente, o intestino delgado dessas aves estava mais esbranquiçado, com conteúdo aquoso, mucoso ou espumoso no lúmen, além de alimento pobremente digerido. Histologicamente, havia enterite com necrose e dilatação discreta a acentuada das criptas com achatamento do epitélio. O jejuno foi o segmento mais acometido histologicamente. A morfometria revelou atrofia das vilosidades especialmente no jejuno e íleo. Alterações histológicas em outros órgãos não foram observadas. O sequenciamento de última geração (NGS) detectou vírus da nefrite aviária (ANV), rotavírus aviário (AvRV) e picornavírus em jejuno dos pintinhos de sete dias. Em pintinhos de um dia, nenhum vírus foi detectado no jejuno. A análise da sequência molecular do picornavírus revelou similaridade genética com o gênero Gallivírus (GV). No teste exato de Fisher, somente o GV foi estatisticamente significativo (p<0,05), comparando pintinhos com e sem lesão histológica. Hibridização in situ (ISH) para GV foi realizada e associação entre o vírus e a lesão não foi encontrada, ou seja, este vírus estava presente somente em células de criptas sem lesão histológica, embora adjacentes a estas. Pintinhos, incluindo aqueles negativos no NGS e sem lesões no jejuno, apresentaram marcação para GV na ISH, apesar desta marcação ter sido em menor número quando comparada aos animais com lesão no jejuno. Apesar da exata etiologia da RSS não ser bem compreendida e vários vírus terem sido descritos, os resultados sugerem que o GV pode desempenhar algum papel na patogênese da RSS em associação com outros vírus, como ANV e AvRV.Runting Stunting Syndrome (RSS) in commercial chickens has been reported worldwide and, although several studies have been carried out to clarify the etiology, there are some gaps in relation to the epidemiological, pathogenic and etiological aspects. In order to characterize the intestinal lesions in the different segments and age from diseased chicks, in addition to investigating the etiology of RSS, naturally infected chicks presenting growth retardation, apathy, poorly digested food in the feces or diarrhea were collected from four different commercial farms located in three municipalities in the in the state of Minas Gerais. Chicks without clinical signs were used as control. The total number of examined chicks were 243. The average weight of affected chicks was lower than expected for their flocks. Macroscopically, the small intestine of these chicks had paler serosa, with a watery, mucous or foamy content in the lumen associated with poorly digested food. Histologically, there was mild to marked dilation of the crypt with flattening of the crypt epithelium and necrotic crypts. Jejunum was the segment more affected histologically. The histologic morphometry of the intestines revealed villous atrophy especially in jejunum and ileum. Histological changes in other organs were not observed. Next-generation sequencing (NGS) detected avian nephritis virus (ANV), avian rotavirus (AvRV) and picornavirus in jejunum from 7-days-old chicks. In the one-day-old chicks no viruses were detected in the jejunum. Molecular sequence analysis of the picornavirus revealed genetic similarity with Gallivirus (GV) genus. In Fisher's exact test, only the GV was statistically significant (p <0.05), comparing chicks with and without histological lesions. In situ hybridization (ISH) for GV was performed and no association between virus and lesion was found, in other words, this virus was present only in crypt cells without histological lesion, although adjacent to them. Chicks, including those negative in NGS and without jejunal lesions, presented ISH positive signals for GV, in spite of low number of label in comparison with animals with jejunal lesions.
