No presente estudo, avaliamos a participação da angiotensina-(1-7) [Ang-(1-7)] receptor Mas nos efeitos benéficos do treinamento aeróbio em um modelo experimental de asma. Camundongos Balb/c foram submetidos a um protocolo crônico de asma (sensibilização com OVA), infundidos cronicamente com o antagonista do receptor Mas, o A779 (1µg/h), e treinados 5x/semana (moderada intensidade/esteira). Os níveis séricos de IgE foram determinados por ELISA, o remodelamento das vias aéreas por histologia, a infiltração de células inflamatórias por ensaio de atividade enzimática. Além disso, o desempenho físico foi determinado por teste de esforço máximo. A expressão gênica do PGC1-α foi determidada por qRT-PCR, a expressão proteica do receptor Mas e de fibronectina foi determinada por Western blotting. Os principais resultados obtidos foram, o tratamento com A779 impediu a redução dos níveis séricos de IgE, a redução da deposição de matriz extracelular nas vias aéreas, a redução do espessamento da parede alveolar e a redução da atividade da enzima MPO nos animais asmáticos treinados. A expressão proteica do receptor Mas no pulmão não foi alterada em nenhum grupo experimental. O treinamento aeróbio foi confirmado pelo aumento da expressão gênica de PGC1-α nos animais controle e asmáticos. Estes dados sugerem que a ativação do receptor Mas está envolvida, pelo menos em parte, nos efeitos anti-inflamatórios e anti-fibróticos induzidos pelo treinamento aeróbio de moderada intensidade em animais submetidos a um modelo experimental crônico de asma.Asthma is a chronic inflammatory airway disease that involves multiple cells in particular T-lymphocytes, mast cells, eosinophils, neutrophils and epithelial cells. In the present study, we evaluated the participation of the angiotensin-(1-7) [Ang-(1-7)] receptor Mas, in the beneficial effects of aerobic training in an experimental model of asthma. Balb/ c mice were subjected to a protocol chronic asthma (OVA sensitization), infused with the Mas receptor antagonist, A779 (1µg/h), and trained 5x/week (moderate/ treadmill). The serum IgE levels were determined by ELISA, the airway remodelling by histology, infiltration of inflammatory cells by enzyme activity assay. Furthermore, the physical performance was determined by maximum effort test. The gene expression of PGC-1 α was determined by qRT-PCR, protein expression of the Mas receptor and fibronectin by Western blotting. The main results were, treatment of asthmatic and trained animals with A779 prevented the reduction in serum IgE levels, reduction in extracellular matrix deposition in the airways, reduction in the alveolar wall thickening, reduction in MPO enzyme activity. Protein expression of the Mas receptor in the lung was not altered. Physical training was confirmed by the increase in PGC1-α in control or asthmatic animals.
