Currently,in situdiagnosis is determined through anamnesis, evaluation ofthe history, and perception of the clinical signs of the herd. There is no practical andnoninvasive test adapted to a large number of samples, which represents a gap forthe use of new approaches that provide information about infections and the risksof herds. Here, we performed a search in theNeospora caninumgenome by linearB-cell epitopes using immunoinformatic tools aiming to develop a chimeric proteinwith high potential to bind specifically to antibodies from infected cattle samples.An enzyme-linked immunosorbent assay with the new chimeric antigen was devel-oped and tested with sera from natural fieldN. caninum-infected bovines. The cross-reactivity of the new antigen was also evaluated using sera from bovines infected byother abortive pathogens, includingTrypanosoma vivax,Leptospirasp.,Mycobacte-rium bovis, andBrucella abortus, and enzootic bovine leucosis caused by bovine leu-kemia virus, as well as with samples of animals infected withToxoplasma gondii. Theassay using the chimeric protein showed 96.6% 3.4% of sensitivity in comparisonto healthy animal sera. Meanwhile, in relation to false-positive results provided bycross-reactivity with others pathogens, the specificity value was 97.0% 2.9%. Inconclusion, immunoinformatic tools provide an efficient platform to build an accu-rate protein to diagnose bovine neosporosis based on serum samplesA neosporose tornou-se uma preocupação, uma vez que está associada ao aborto em bovinos. Atualmente, o diagnóstico in situ é determinado por meio de anamnese, avaliação da história e percepção dos sinais clínicos do rebanho. Não existe um teste prático e não invasivo adaptado a um grande número de amostras, o que representa uma lacuna para a utilização de novas abordagens que forneçam informações sobre infecções e riscos dos rebanhos. Aqui, realizamos uma pesquisa no genoma de Neospora caninum por epítopos lineares de células B usando ferramentas de imunoinformática com o objetivo de desenvolver uma proteína quimérica com alto potencial para se ligar especificamente a anticorpos de amostras de bovinos infectados. e testado com soros de campo naturalN. bovinos infectados com caninum. A reatividade cruzada do novo antígeno também foi avaliada utilizando soros de bovinos infectados por outros patógenos abortivos, incluindo Trypanosoma vivax, Leptospirasp., Mycobacte-rium bovis e Brucella abortus, e leucose enzoótica bovina causada pelo vírus da leucemia bovina, bem como com amostras de animais infectados com Toxoplasma gondii.
