Caracterização fenotípica e genotípica de espécies de leveduras do complexo Cryptococcus isoladas de amostras da poeira de bibliotecas de Cuiabá e Várzea Grande e sua relação com árvores hospedeiras

Resumo: | The Cryptococcus spp complex is composed of cosmopolitan distribution capped yeasts, including etiological agents of cryptococcosis. In the environment the fungus is found mainly in substrates of animal and vegetal origins. Human infection occurs through the inhalation of sexually propagated propagules present in the environment. In this work 42 samples were collected with the help of swab, gravitational exposure and Andersen sampler in dust found in the books belonging to the collection of seven libraries in the city of Cuiabá and Várzea Grande / Mato Grosso. The collected material was sown on Sabouraud agar, and after observing the characteristics compatible with colonies of creamy to mucoid aspect, they were isolated in Niger agar and cultured at temperatures of 27º and 37oC for 5-7 days. Identification of the isolated samples was performed by microscopic observation on fresh preparations stained by China ink, additional tests on CGB (L-Canavanine-Bromothymol blue-glycine), urea broth and carbohydrate assimilation tests (auxanogram) and Posterior molecular identification was performed in a specialized laboratory. Of the 42 samples collected from the dust of books, 1,306 (70%) of colonies were registered in libraries in Cuiabá and 559 (30%) in the libraries in Várzea Grande, of positive samples totaling 1,865 CFUs. The most frequently isolated species was C. gattii 46 (2.5%); Followed by C. neoformans 28 (1.5%, C. terreus 15 (0.8%), C. luteolus 12 (0.6%) and C. unigutullatus (0.5%), the remaining 94% are identified. The high biodiversity of yeasts of the genus Cryptococcus spp, isolated from several environmental sources in urban areas of Brazil, suggests the possibility of immunocompromised and even healthy individuals coming into contact with multiple sources of contagion daily throughout their lives This study points to the possible acquisition of the isolated dust fungus in library collections, due to the continuous exposure of the employees working in these environments and individuals visiting these places to the microniche present in the librarian environment. |

No meio ambiente o fungo é encontrado principalmente em substratos de origens animal e vegetal. A infecção humana ocorre através da inalação de propágulos de origem sexual presentes no ambiente. Neste trabalho foram realizadas 42 coletas com o auxílio de swab, exposição gravitacional e amostrador de Andersen em poeira encontrada nos livros pertencentes ao acervo de sete bibliotecas da cidade de Cuiabá e Várzea Grande/Mato Grosso. O material coletado foi semeado em ágar Sabouraud, e após observação das características compatíveis com colônias de aspecto cremoso a mucóide, as mesmas foram isoladas em ágar niger e cultivadas a temperaturas de 27º e 37oC C por 5-7 dias. A identificação das amostras isoladas foi realizada através da observação microscópica em preparações a fresco coradas pela tinta da China, testes adicionais em meio CGB (L-Canavanina-glicina-azul de bromotimol), caldo uréia e testes para assimilação de carboidratos (auxanograma) e posterior identificação molecular foi realizada em laboratório especializado. Das 42 amostras coletadas da poeira de livros, foram isoladas 1.306 (70%) de colônias em bibliotecas em Cuiabá e 559 (30%) nas bibliotecas em Várzea Grande, de amostras positivas totalizando 1.865 UFC’s. A espécie mais frequentemente isolada foi C. gattii 46 (2,5%); seguido de C. neoformans 28 (1,5%, C. terreus 15 (0,8%), C. luteolus 12 (0,6%) e C. unigutullatus (0,5%) os demais 94% encontram-se em identificação. A alta biodiversidade de leveduras do gênero Cryptococcus spp, isoladas de diversas fontes ambientais nas áreas urbanas do Brasil, sugere a possibilidade de indivíduos imunocomprometidos e até mesmo hígidos entrarem em contato com múltiplas fontes de contágio diariamente ao longo de suas vidas.
