Análise fractal e modelos matemáticos para a investigação da inativação fototérmica de Candida albicans usando nanotubos de carbono

Resumo: | Candida albicans is responsible for the majority of nosocomial infections affecting immunocompromised patients. Systemic antifungals may promote microbial resistance, which has led to the search for alternative treatments, such as photothermal therapy (PTT). PTT assumes that the interaction of electromagnetic radiation with a photothermal agent generates heat that can lead to the destruction of tumor cells and the death of microorganisms. Carbon nanotubes (CNTs) have the potential for applications in biomedical systems, including acting as controlled deliverers of drugs, biosensors and scaffolds for tissue engineering and regenerative medicine. Furthermore, the absorption of radiation by CNTs in the infrared region induces an increase in temperature, which makes CNTs candidates for photothermal agents. In this work, the photothermal inactivation of C. albicans was evaluated by multiple wall CNTs associated with laser radiation in the near-infrared region. The mechanisms that are involved in inactivation were evaluated through cell susceptibility studies and an analysis of microscopic images that are associated with mathematical models and fractal concepts. The results indicate that direct contact between the cells and CNTs without irradiation does not lead to cell death, whereas the laser-mediated process is effective in inactivation. The application of the laws of scale and fractal concepts indicate that in the control groups, there are two distinct regimes that are delimited by the mean diameter of the microorganisms, as described by the Eden model and by the quasi-Euclidean surface. For the irradiated groups, the surfaces present only one regime described by KPZ. The analysis of the fractality of the system by mathematical models can help in the identification of new strategies for the inactivation of microorganisms. |

Antifúngicos sistêmicos podem promover a resistência microbiana, e isto tem levado a busca por tratamentos alternativos, tal como a terapia fototérmica, (PTT). A PTT parte da premissa de que a interação da radiação eletromagnética, geralmente radiação infravermelha, com um agente fototérmico gera aquecimento que pode levar à destruição de células tumorais e morte de microrganismos. Nanotubos de carbono, (CNTs), são uma classe de nanomateriais que têm potencial para aplicações em sistemas biomédicos, incluindo entrega controlada de fármacos, biossensores e scaffolds para engenharia de tecidos e medicina regenerativa. Além disso, por absorverem radiação na região do infravermelho, induzindo aumento da temperatura, são candidatos a agentes fototérmicos. Neste trabalho, a inativação fototérmica da C. albicans foi avaliada usando CNTs de múltiplas paredes associados à radiação laser no infravermelho próximo. Os mecanismos envolvidos na inativação, foram avaliados através de estudos de susceptibilidade celular e análise de imagens microscópicas associadas a modelos matemáticos e conceitos fractais. Os resultados indicam que o contato direto entre as células fúngicas e os CNTs sem irradiação, não levam à morte celular, ao passo que o processo mediado por laser é eficaz na inativação, que pode ter sido causada pela ruptura das paredes celulares. A aplicação das leis de escala e conceitos fractais indicam que nos grupos controles há dois regimes distintos delimitados pelo diâmetro médio do microrganismo. Para comprimentos maiores que esse diâmetro a superfície é descrita pelo modelo de Eden e, para comprimentos menores a superfície é quase-Euclidiana. Para os grupos irradiados de C. albicans + CNTs as superfícies apresentam apenas um regime, com fractais self-affine descritos por KPZ (Kardar-Parisi-Zhang).
