Prevalência e diversidade genética de Trypanosoma theileri em bovinos leiteiros de Ji-Paraná, Rondônia

Resumo: Trypanosoma (Megatrypanum) theileri is a flagellated protozoan that is infectious for ruminants from different countries. Through this study we investigated the prevalence of the agent in cattle from 64 farms of family farms, divided into 06 rural sectors in accordance with Technical Assistance and Rural Extension Association of the State of Rondônia. We also analyzed the genetic diversity of T. theileri isolates of cattle in the municipalities of Ji-Paraná. DNA from 320 blood samples and 13 isolates of T. theileri from cows underwent diagnostic PCR based on amplification of the Cathepsin L-like protein (CATL) gene and glycosomal glyceraldehyde genes 3- phosphate dehydrogenase (gGAPDH) and a small subunit rRNA (V7V8 SSU rRNA) region only for isolates. The prevalence in cattle was 8,13% (95% IC: 6,18%- 10,59%), demonstrating the T. theileri circulation in the municipality. Then with phylogenetic analyzes with the amplicons of CATL, gGAPDH and V7V8 SSU rRNA genes we verified the first time the presence of genotypes TthIB in catlle from western Amazon and TthIF in cattle from Brazil. Indeed, we identified a possible new genotype TthIL de T. theileri.

Por meio desse estudo foi investigada a prevalência do agente em bovinos de 64 propriedades rurais de agricultura familiar, divididas em 06 setores rurais de acordo com Associação de Assistência Técnica e Extensão Rural do Estado de Rondônia. Nós também analisamos a diversidade genética de isolados de T. theileri de bovinos do município. DNA de 320 amostras de sangue e 13 isolados de T. theileri foram submetidos a PCR diagnóstico baseada na amplificação do gene Catepsina L-“like” (CATL) e do gliceraldeído fosfato desidrogenase (gGAPDH) e V7-V8 SSUrDNA somente para os isolados. A prevalência no gado foi de 8,13% (95% IC: 6,18%-10,59%) demonstrando a circulação do T. theileri no município. Além disso, por meio de análises filogenéticas com amplicons dos genes CATL, gGAPDH e V7V8 SSUrDNA, nós verificamos a presença do genótipos TthIB em bovinos da Amazônia ocidental e TthIF em bovinos no Brazil pela primeira vez.
