Resumo: Breast cancer is one of the most frequent diseases among women, and now the search for more effective treatments is still a challenge. In this sense the development of drugs and / or herbal compounds has increased in recent years and the combination of active ingredients modified release systems have been used to enhance or stimulate immune responses. Medicinal plants have always been objects of study in an attempt to find new sources of active ingredients. Among them stands out the Herbal Mixture compound having modulatory activity can activate the immune system. This compound adsorbed to the microsphere polyethylene glycol was able to increase the functional activity of human phagocytes is important in bacterial infections, but its effects on tumor cells has not yet been elucidated. This work aimed to evaluate the effects of the compound Herbal Mixture adsorbed polyethylene glycol microsphere in tumor breast cell lines (MCF-7) in co-culture with mononuclear cells blood. Were analyzed in the presence of compound Herbal Mixture adsorbed microsphere polyethylene glycol blood viscosity, as well as the inhibition of proliferation, apoptosis, cell viability, release of intracellular calcium mononuclear cells in coculture with mammary adenocarcinoma cells (MCF-7). It was found that compound Herbal Mixture does not change the viscosity of normal blood. When we assessed cell viability in the presence of compound Herbal Mixture polyethylene glycol adsorbed microsphere was observed that the viability rate after 24 hours was 95.55% for mononuclear cells and 40.19% for MCF-7 cells. An increase of apoptosis and intracellular calcium in MCF-7 cells in the presence of compound Herbal Mixture. The co cell culture mononuclear and MCF-7 in presence of compound Herbal Mixture polyethylene glycol adsorbed to reduced cell viability, increased apoptosis rates and release of intracellular calcium. These data suggest that compound Herbal Mixture absorbed microsphere polyethylene glycol can act as functional material with antitumor activity against MCF-7 cells.

Nos últimos anos o desenvolvimento de fármacos e/ou compostos fitoterápicos associados a sistemas de liberação modificada vem sendo utilizados para potencializar ou estimular respostas imunológicas. As plantas medicinais sempre foram objetos de estudos na tentativa de descobrir novas fontes de princípios ativos, entre elas destaca-se o composto “Mais Vida” que apresenta atividade moduladora sobre o sistema imunológico. Este composto adsorvido a microesfera de polietilenoglicol aumenta a atividade funcional de fagócitos humanos sendo importante em infecções bacterianas, porém seus efeitos sobre células tumorais ainda não foram elucidados. Assim, este trabalho teve como objetivo avaliar os efeitos do composto “Mais Vida” adsorvida a microesfera de polietilenoglicol em linhagens de células tumorais de mama (MCF-7), e em cocultura com células mononucleares do sangue humano. Foram analisadas na presença do composto “Mais Vida” adsorvido a microesfera de polietilenoglicol a viscosidade sanguínea, a viabilidade celular, a inibição de proliferação, apoptose e liberação de cálcio intracelular de células em cocultura com células de adenocarcinoma mamário (MCF-7). Verificou-se que o composto “Mais Vida” não alterou a viscosidade sanguínea. Quando se avaliou a viabilidade celular na presença do composto “Mais Vida” adsorvido a microesfera de PEG observou-se que o índice de viabilidade após 24 horas foi de 95,55% para células mononucleares e de 40,19% para células MCF-7. Houve aumento da apoptose e cálcio intracelular em células MCF-7 na presença do composto “Mais Vida”. A cocultura de células mononucleares e MCF-7 em presença do composto “Mais Vida” adsorvido ao polietilenoglicol reduziu a viabilidade celular, aumentou os índices de apoptose e a liberação de cálcio intracelular.
