Efeitos de nanodoses de Bário adsorvidos a microesferas de polietilenoglicol (PEG) sobre cocultura de células mononucleares do sangue humano e linhagens de células de câncer de mama (MCF-7)

Resumo: | In recent decades due to the increasing number of diseases related to heavy metals, there has been great interest in the biological effects of these compounds. Among these heavy metals, Barium (Ba) is highly toxic because it has a high potential for absorption by the organism. Based on this need to improve human health on exposure to contaminants, systems have been invested in modulating immune responses Several works have shown the associated use of PEG microspheres with chemical compounds to act in the treatment of infections and some diseases, such as cancer. On the other hand, barium has been shown to be a potent toxic agent for human cells, causing high rates of apoptosis. It is possible that adsorption of barium to PEG microspheres may interfere with the toxic actions of barium. Thus, this work aimed to evaluate the effects of barium nanodegradation adsorbed on polyethylene glycol (PEG) microspheres in human blood mononuclear cells and breast cancer cells (MCF-7). The adsorbed barium PEG microspheres were characterized and their effects on blood viscosity, cell viability and apoptosis were verified. The PEG microspheres were shown to have spherical, stable structures capable of adsorbing, regardless of the dose, BaCl2 and with a size corresponding to 7,0 μm. It was found that barium nanodoses did not alter the viscosity of normal blood. When evaluating cell viability in the presence of different doses of barium, a significant difference was observed in MCF-7 cells with a higher index for BaCl2 25pg/L and a reduction in cell viability when in coculture, especially when the cells were treated with BaCl2 1ng/L. There was an increase in apoptosis of cells incubated with BaCl2 1ng/L adsorbed to PEG microspheres, and the highest apoptosis index (85.99%) was observed in cell culture. These data suggest that BaCl2 1ng/L adsorbed to PEG microspheres may act as functional material with antitumor activity on MCF-7 cells. |

Dentre estes metais pesados apresenta-se o Bário (Ba) que é altamente tóxico, pois apresenta potencial elevado de absorção pelo organismo. Baseado nesta necessidade de melhorar a saúde humana quanto à exposição aos contaminantes tem se investido em sistemas capazes de modular as respostas imunológicas. Diversos trabalhos têm mostrado o uso associado das microesferas de PEG com compostos químicos de modo a atuarem no tratamento de infecções e algumas doenças, tais como o câncer. Por outro lado, o bário tem se mostrado um potente agente tóxico para células humanas, causando altos índices de apoptose. É possível que a adsorção do bário a microesferas de PEG possa interferir nas ações tóxicas do bário. Assim, este trabalho teve como objetivo avaliar efeitos de nanodoses de bário adsorvido a microesferas de polietilenoglicol (PEG) em células mononucleares do sangue humano e em células do câncer de mama (MCF-7). Foram caracterizadas as microesferas de PEG com bário adsorvido e verificado seus efeitos sobre a viscosidade sanguínea, a viabilidade celular, a apoptose. As microesferas de PEG se mostraram com estruturas esféricas, estáveis capazes de adsorver, independente da dose, o BaCl2 e com tamanho correspondente a 7,0 µm. Verificouse que as nanodoses de Bário não alteraram a viscosidade do sangue normal. Quando se avaliou a viabilidade celular na presença das diferentes doses de Bário verificou-se diferença significativa nas células MCF-7 com maior índice para BaCl2 25pg/L e redução da viabilidade celular quando em cocultura, principalmente quando as células foram tratadas com BaCl2 1ng/L. Houve aumento na apoptose de células incubadas com BaCl2 1ng/L adsorvido a microesferas de PEG, sendo que o maior índice de apoptose (85,99%) foi observado em cocultura celular.
